摘要
目的:通过生物信息学途径探究过氧化物还原酶4(peroxiredoxin 4,PRDX4)在胃癌(gastric cancer,GC)中的表达及其意义,并通过免疫组织化学检测63例GC患者肿瘤组织中PRDX4的表达情况。方法:从Ualcan数据库获取正常胃组织及原发性胃癌组织PRDX4表达数据,在LinkedOmics中检索PRDX4的共表达基因,并通过GO富集分析、KEGG通路分析确定这些基因参与的生物学过程及功能,利用STRING数据库分析相关基因编码蛋白的互作网络。最后利用GEPIA数据库分析PRDX4表达与胃癌患者预后的关系;纳入2022年1月至2022年10月在成都医学院第一附属医院就诊的63例GC患者,通过MaxVision法对GC肿瘤组织及其周围正常组织进行PRDX4表达情况的免疫组化染色并观察结果。结果:Ualcan数据库显示,与周围正常胃组织相比,PRDX4在GC组织中高度表达(P<0.05),但尚未发现其表达与性别、肿瘤分化程度、肿瘤大小和淋巴结转移有关联。同时检索出PRDX4共表达基因主要与生物调节、代谢过程、刺激反应等分子生物学过程存在紧密联系,并与ErbB、Hippo、Hedgehog等信号通路及各种癌症通路显著相关。多个GC相关基因编码蛋白与PRDX4所编码蛋白存在互作关系。经免疫组化分析,PRDX4在63例GC患者的GC组织中高度表达。结论:PRDX4在GC组织中高表达,且与GC的发生和发展密切相关,有望成为GC筛选的一项标志物。
Objective:To explore the expression and significance of PRDX4 in gastric cancer(GC)through bioinformatics,and to detect the expression of PRDX4 in tumor tissues from 63 GC patients with immunohistochemistry.Methods:The expression data of PRDX4 in normal gastric tissues and primary gastric cancer tissues were obtained from Ualcan database,the co-expression genes of PRDX4 were searched on LinkedOmics,and the biological processes and functions of these genes were determined by GO enrichment analysis and KEGG pathway analysis.The interaction network of proteins encoded by related genes was analyzed using STRING database.Finally,GEPIA database was used to analyze the relationship between PRDX4 expression and prognosis of gastric cancer patients.A total of 63 GC patients treated in the First Affiliated Hospital of Chengdu Medical College from January 2022 to October 2022 were included.Immunohistochemical staining of GC tumor tissues and surrounding normal tissues was performed by MaxVision method and the results were observed.Results:Ualcan database showed that PRDX4 was highly expressed in GC tissues compared with surrounding normal gastric tissues,but no correlation was found between PRDX4 and sex,degree of tumor differentiation,tumor size and lymph node metastasis.Meanwhile,PRDX4 co-expression gene was closely related to molecular biological processes such as biological regulation,metabolic process and stimulus response,and was significantly related to ErbB,Hippo,Hedgehog and other signaling pathways and various cancer pathways.Several GC-related gene encoded proteins interacted with PRDX4 encoded proteins.Immunohistochemical and western blot analysis showed that PRDX4 was highly expressed in GC tissues.Conclusion:PRDX4 is highly expressed in GC tissues and is closely related to the ongenesis and development of GC,which is expected to be a marker for GC screening.
