斯氏珀蝽和青革土蝽的线粒体基因组及系统发育分析

摘要 【目的】分析斯氏珀蝽和青革土蝽的线粒体基因组特征及蝽总科的系统发育关系,为更好地理解蝽总科下科级和种级水平的亲缘关系提供依据。【方法】通过测序分析了斯氏珀蝽和青革土蝽的线粒体全基因组数据,并基于此,以已经公布线粒体基因组数据的蝽总科的113个物种和本研究的2种昆虫为内群,2种红蝽总科昆虫为外群,分别利用最大似然法和贝叶斯法构建系统发育树。【结果】斯氏珀蝽和青革土蝽的线粒体基因组均包含37个基因(13个蛋白质编码基因、2个核糖体RNA基因和22个转运RNA基因)和1个非编码控制区;两种昆虫的线粒体基因组全长分别为15346、14632 bp(GenBank序列号:OP919327和OP930890)。两种方法构建的系统发育树一致支持蝽科、龟蝽科、盾蝽科、兜蝽科、荔蝽科、土蝽科、同蝽科、异蝽科和Megarididae均为单系群;斯氏珀蝽与珀蝽为姐妹群,青革土蝽与Macroscytus subaeneus为姐妹群。【结论】斯氏珀蝽与珀蝽亲缘关系最近,青革土蝽与Macroscytus subaeneus亲缘关系最近。 【Objective】Analysis of the mitochondrial genomes of Plautia stali and Macroscytus japonensis and the phylogenetic relationship of Pentatomoidea will provide reference for the understanding on the phylogenetic relationship at the family and species levels of Pentatomoidea.【Method】The mitochondrial genomes of P.stali and M.japonensis were analyzed by sequencing,and based on which the phylogenetic trees were constructed by maximum likelihood and Bayesian inference methods,with 115 Pentatomoidea species as ingroups and 2 Pyrrhocoroidea species as outgroups.【Result】The mitochondrial genomes of P.stali and M.japonensis(GenBank accession No.OP919327 and OP930890)were 15346 bp and 14632 bp in size,and both contained 13 protein-coding genes,2 ribosomal RNA genes,22 transfer RNA genes and a non-coding control region.The phylogenetic trees constructed by both methods shared a similar topological structure,in which Pentatomidae,Plataspidae,Scutelleridae,Dinidoridae,Tessaratomidae,Cydnidae,Acanthosomatidae,Urostylididae and Megarididae were all monophyletic groups;P.stali was sister to P.fimbriata,and M.japonensis was sister to M.subaeneus.【Conclusion】P.staliwas and P.fimbriata share the closest genetic relationship,M.japonensis and M.subaeneus share the closest genetic relationship.
