摘要
单线态氧(^(1)O_(2))是一种重要的高活性氧分子,在生理学和病理过程中起着关键作用。本文构建了^(1)O_(2)响应的基于光诱导电子转移(PET)机理的比率型荧光探针Com-Nap。该探针以扩环香豆素和萘酰亚胺衍生物为荧光团,以蒽和扩环香豆素为识别基团,同时以蒽作为萘酰亚胺荧光团的猝灭基团。单独的探针仅表现出扩环香豆素的荧光信号。当^(1)O_(2)与探针反应后,PET过程被禁阻,呈现出比率型响应信号。该探针实现了对^(1)O_(2)的准确、快速及高选择性检测。
Singlet oxygen(^(1)O_(2)),a biological reactive oxygen species,has recently been found to possess unique biological activity and pharmacology applications.A PET-based ratiometric fluorescent probe Com-Nap for detecting ^(1)O_(2)was developed.A coumarin derivative and a naphthalimide fluorophore were served as the reporters,and 9-methylanthracene was chosen not only as an electron acceptor of PET process(a fluorescence quencher)for naphthalimide but also as recognition unit for ^(1)O_(2)with coumarin derivative(another recognition unit).In the absence of ^(1)O_(2),the probe only showed the fluorescence signal of the coumarin derivative.After the reaction between ^(1)O_(2)and the probe,the PET process was prohibited,which allowed a ratiometric fluorescence response for the detection of ^(1)O_(2).This probe could rapidly detection of ^(1)O_(2)with high selectivity.
