正交设计优选酶解辅助——超声提取花榈木叶总黄酮研究

Optimization of Enzymolysis Assisted Ultrasonic Extraction of Total Flavonoids from leaves of Ormosia henryi by Orthogonal Design

摘要 为研究花榈木叶总黄酮提取工艺,以单因素考察实验为基础,并建立以提取时间(A)为10 min、20 min、30 min,乙醇浓度(B)为60%、70%、80%,液料比(C)为1∶20、1∶25、1∶30,提取次数(D)为1、2、3次的四因素三水平正交实验,以期得到花榈木总黄酮酶解辅助-超声提取最佳工艺。研究得出:花榈木总黄酮酶解辅助-超声提取最优方法为:D_(3)B_(3)C_(3)A_(2),即酶解时间20 min,80%乙醇,液料比1∶30,提取3次。本实验优选出的最佳提取方法方便、可靠、提取率高,适合花榈木总黄酮提取。 In order to study the extraction process of total flavonoids from the leaves of Ormosia henryi,based on the single factor investigation experiment,and orthogonal experiment of four factors and three levels was established with the extraction time(A)as 10 min,20 min,30 min,ethanol concentration(B)as 60%,70%,80%,liquid material ratio(C)as 1∶20,1∶25,1∶30,and extraction times(D)as 1,2,and 3 times,in order to obtain the best technology of enzymatic hydrolysis assisted ultrasonic extraction of total flavonoids from Ormosia henryi.The results showed that the optimal method of enzymatic hydrolysis assisted ultrasonic extraction of total flavonoids from Ormosia henryi was D_(3)B_(3)C_(3)A_(2):enzymatic hydrolysis time was 20 min,80%ethanol,liquid to material ratio was 1∶30,and extraction was conducted for three times.The best extraction method selected in this experiment was convenient,reliable and has high extraction rate,which was suitable for the extraction of total flavones from Palm.
