摘要
该文对CTIF在胚胎发育过程中的表达进行初步研究。通过设计特异性扩增W拷贝、Z拷贝以及同时扩增W和Z拷贝的引物(CTIF-W、CTIF-Z及CTIF-ZW),对其在胚胎的不同日龄的组织的mRNA表达量进行分析,并通过FAD(fadrozole)诱导性反转探索其对性别表型的响应。结果显示,CTIF-W在孵化6 d的胚胎性腺中的表达高于尾部,且随胚胎发育表达量逐渐升高,而CTIF-Z和CTIF-ZW的表达趋势相似;而在6 d鸡胚的其他组织中,CTIF-ZW在肾中的表达显著高于其他组织,说明CTIF在发挥某些基础功能的同时可能还与肾脏的发育或功能相关。CTIF-Z在多数组织中表达量均呈现出雄性高于雌性,而CTIF-ZW在雌雄之间的表达量差异则相对较小,表明W拷贝可能在一定程度上调控了雌性中Z拷贝的表达。CTIF-W和CTIF-Z在12 d的胚胎组织中均表达,且均高度表达于脾脏组织中;CTIF-Z在大多数组织中雄性表达高于雌性。此外,CTIF-W表达受雌性向雄性性反转的影响而显著降低,而CTIF-Z则没有响应。综上,CTIF在鸡胚的肾脏和脾脏中高度表达,且其Z拷贝在雌雄组织之间存在剂量效应,而W拷贝可能在雌性中作为Z拷贝的补偿以保证其生物学功能的发挥。
In this paper,the expression of CTIF during embryonic development was studied.By designing primers(CTIF-W,CTIF-Z and CTIF-ZW)that specifically amplify W and Z copies and simultaneously amplify W and Z copies,the mRNA expression levels of W and Z copies in different day-old tissues of embryos were analyzed,and the response to gender phenotype was explored by FAD(fadrozole)induced reversal.The results showed that the expression of CTIF-W in the gonads of embryos at 6 days of incubation was higher than that in the tail,and the expression level gradually increased with embryonic development,while the expression trend of CTIF-Z and CTIF-ZW was similar.In other tissues of 6-day-old chicken embryos,the expression of CTIF-ZW in kidney was significantly higher than that in other tissues,indicating that CTIF may also be related to the development or function of kidney while exerting certain basic functions.The expression of CTIF-Z in most tissues was higher in males than in females,while the expression difference of CTIF-ZW between males and females was relatively small,indicating that W copy may regulate the expression of Z copy in females to a certain extent.Both CTIF-W and CTIF-Z were expressed in 12-day embryonic tissues and were highly expressed in spleen tissues.The expression of CTIF-Z in male was higher than that in female in most tissues.In addition,the expression of CTIF-W was significantly reduced by the effect of female-to-male sex reversal,while CTIF-Z did not respond.In summary,CTIF is highly expressed in the kidney and spleen of chicken embryos,and its Z copy has a dose effect between male and female tissues,while W copy may be used as a compensation for Z copy in female to ensure its biological function.
