基于EST-SSR分子标记的萱草亲缘关系分析

摘要 萱草是极具观赏价值的多年生宿根草本植物,在我国具有悠久的栽种历史。为了探明36个萱草品种间的亲缘关系,利用毛细管电泳荧光标记技术对36个萱草品种进行了EST-SSR分子标记检测,利用11对多态性良好的引物对36份供试萱草材料进行PCR扩增。结果显示:11对引物共获得40个多态性位点,样本平均有效等位基因数为2.58个,平均Shannon多样性指数为1.00,平均期望杂合度为0.57,平均观察杂合度为0.52;以遗传距离矩阵为分析对象,36个萱草品种间的遗传距离为0.05~0.70,利用NTSYS软件按UPGMA法聚类结果显示,遗传距离0.48为阈值时,36种萱草品种分为2个大类,遗传距离0.414为阈值时,可进一步将第1大类分为4个亚类。综上,供试的36个萱草品种之间具有较高的遗传多样性和较为丰富的遗传背景,为今后培养萱草优良新品种提供了一定的理论依据。 Hemerocallis fulvay is a perennial hosta with great ornamental value and has a long history of cultivation in China.In order to explore the kinship relationship among 36 Hemerocallis fulvay species.In this study,EST-SSR molecular marker detection of 36 Hemerocallis fulvay species was carried out by capillary electrophoresis fluorescence labelling technique,and PCR amplification of 36 test Hemerocallis fulvay materials was carried out using 11 pairs of primers with good polymorphism.The results showed that a total of 40 polymorphic loci were obtained from the 11 primer pairs,with an average number of 2.58 effective alleles in the samples,an average Shannon diversity index of 1.00,an average expected heterozygosity of 0.57,and an average observed heterozygosity of 0.52.Analysed by the genetic distance matrix,the genetic distances among the 36 Hemerocallis fulvay varieties ranged from 0.05 to 0.70,the clustering results by UPGMA method using NTSYS software showed that the 36 Hemerocallis fulvay varieties were classified into two major classes when the genetic distance of 0.48 was the threshold,and the first major class could be further divided into four subclasses when the genetic distance of 0.414 was the threshold.In conclusion,the 36 Hemerocallis fulvay varieties for testing have high genetic diversity and richer genetic backgrounds,which provides a certain theoretical basis for cultivating excellent new varieties of Hemerocallis fulvay in the future.
