地龙对糖尿病肾病大鼠肝脏和肾脏保护作用的研究

摘要 探讨中药地龙水煎剂对高糖高脂联合链脲佐菌素(STZ)诱导糖尿病肾病(DN)大鼠肝脏和肾脏的保护作用及其机制。将健康60只SD大鼠随机分为6组:正常组、模型组和地龙水煎液高、中、低剂量组及阳性对照组,每组10只。每日观察各组动物的一般情况;药物干预4周后,腹主动脉取血,测定各组动物全血RBC、PLT、WBC、Hb计数,血清低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、总胆固醇(TC)、甘油三酯(TG)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、空腹血糖(FBG)、血肌酐(SCr)、尿素氮(BUN)、血尿酸(UA)等指标;取各组动物右肾,测定肾重/体质量。取各组动物肝左叶和左肾,HE染色后行肝组织、肾组织病理学观察。WBC、RBC、PLT、LDL-C、HDL-C、TC、TG、BUN、UA、SCr、FBG计数,模型组高于正常组,地龙各药物组低于模型组(P<0.05);地龙药物组与贝那普利组相比,无显著差异。Hb模型组低于正常组;地龙各药物组高于模型组;组织切片结果显示,中、高剂量地龙水煎剂、贝那普利组可以改善糖尿病肾病大鼠肝纤维化程度;肾脏系膜基质、管型、颗粒变性等程度。地龙水煎剂能够改善DN大鼠的肝脏和肾脏病变,降低DN大鼠的TG、TC、HDL-C、ALT、AST等指标,降低DN大鼠尿蛋白水平,其机制与地龙有效成分改善动物血糖血脂紊乱状态,抑制炎症反应,保护肝脏肾脏组织有关。 To investigate the protective effect and mechanism of earthworm decoction on liver and kidney of diabetic nephropathy(DN)rats induced by high glucose and high fat combined with streptozotocin(STZ).Sixty healthy SD rats were randomly divided into 6 groups:normal group,model group,high,medium and low dose group of earthworm decoction and positive control group,with 10 rats in each group.Daily monitoring of the overall health and well-being of each animal group is essential;Four weeks after the drug intervention,blood was taken from the abdominal aorta to measure the whole blood RBC,Plt,WBC,Hb counts,serum low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),total cholesterol(TC),triglycerides(TG),alanine aminotransferase(ALT),aspartate aminotransferase(AST),fasting blood glucose(FBG),blood creatinine(SCr),blood urea nitrogen(BUN),blood uric acid(UA)and other indicators.The right kidney of each group was taken and the kidney weight/body weight was measured.The left lobe of liver and left kidney of each group were taken,and the liver tissue and kidney tissue were observed after HE staining.WBC,RBC,RBC,PLT,LDL-C,HDL-C,TC,TG,BUN,UA,SCr,FBG counts of the rats in the model group rose obviously compared with normal control group,Compared with the model group(P<0.05),the drug groups of earthworm were significantly reduced,and there was no statistically significant difference between the earthworm group and the benazepril group.Test results displayed that compared with model control group the Hb counts enhanced in the normal group,the counts enhanced in the earthworm groups.And tissue section results showed that medium and high doses of earthworm decoction and benazepril group could improve liver fibrosis in diabetic nephropathy rats.Degree renal mesangial stroma,molding and granular denaturation.Earthworm decoction can improve the liver and kidney lesions of DN rats,reduce the TG,TC,HDL-C,ALT,AST and other indicators of DN rats,and reduce the urinary protein level of DN rats.The mechanism is related to the improvement of animal blood glucose and lipid disorders,inhibition of inflammatory response,and protection of liver and kidney tissues.
