基于HPLC法测定骨髓炎颗粒中4种主要有效成分含量

摘要 目的建立骨髓炎颗粒中黄芩苷、绿原酸、黄芪甲苷、汉黄芩苷等主成分含量测定方法,为深入研究骨髓炎颗粒奠定基础。方法采用HPLC法。色谱条件:Diamonsil C18色谱柱(200 mm×4.6 mm,5μm);以乙腈-0.1%磷酸溶液为流动相进行梯度洗脱;检测波长280 nm;流速为1.0 mL/min;柱温30℃;进样量10μL。结果绿原酸、黄芪甲苷、黄芩苷、汉黄芩苷分别在质量浓度6.00~120、5.75~115、34.0~680、7.55~151μg/mL范围内线性关系良好,r均大于0.9995;加样回收率分别为97.71%、100.36%、99.81%、101.05%,RSD值均小于2.0%。结论所建立的含量测定方法操作简单,重复性及准确性较好,可用于骨髓炎颗粒的质量标准评价。 Objective To establish the content determination method of baicalin,chlorogenic acid,astragaloside and wogonoside,it would lay a foundation for further study of osteomyelitis granules.Methods HPLC method was used.Chromatographic conditions:Diamonsil C18 column(200 mm×4.6 mm,5μm),the gradient elution was carried out with acetonitrile-0.1%phosphoric acid solution as mobile phase.Detection wavelength 280 nm,the flow rate was 1.0 mL/min,column temperature 30℃,the sample size was 10μL.Results Chlorogenic acid,astragaloside,baicalin and wogonoside were well related linearly within the mass concentration 6.00-120、5.75-115、34.0-680、7.55-151μg/mL,respectively,with r greater than 0.9995.The recovery rates of the added samples were 97.71%,100.36%,99.81%and 101.05%,respectively,and the RSD values were less than 2.0%.Conclusion The established method has the advantages of simple operation,good repeatability and accuracy,and can be used to evaluate the quality standard of osteomyelitis granules.
