摘要
为揭示光合色素影响紫花槭(Acer pseudosieboldianum)叶片呈色的分子机制,挖掘与光合色素密切相关的功能基因。本研究基于紫花槭转录组数据,将检测到的Unigene定位到具体的代谢途径分支,对与光合色素有关的KEGG注释编号(KO)进行汇总,得到卟啉与叶绿素代谢途径(ko00860)达27个途径分支,类胡萝卜素生物合成途径(ko00906)有17个途径分支。从中筛选出66条与光合色素相关的差异表达基因(DEGs),其中28条DEGs表达量上调,38条DEGs表达量下调。对筛选到的光合色素相关途径DEGs进行功能预测,在66条DEGs中共筛选出28条结构基因编码了紫花槭光合色素代谢途径中19个关键酶(POR, DVR, NOL,LCYB, NCED, CYP707A等),具体可划分为四类功能。把筛选到的结构基因与KOG数据库中的蛋白序列进行比对并进行GO功能分类统计,富集到数目最多的是生物过程(biological process, BP),其中与代谢过程、单生物过程和细胞过程相关的功能富集最显著。对光合色素相关基因的筛选有助于揭示紫花槭叶色形成分子机制,为今后研究紫花槭基因定位、基因克隆等提供数据支撑。
In order to reveal the intrinsic molecular mechanis m of photosynthetic pigment in Acer pseudosieboldianum,the functional genes closely related to photosynthetic pigments were explored.Based on the transcriptional data of Acer pseudosieboldianum,we locate the detected Unigene to specific metabolism pathway branches.We summarize KEGG annotation number(KO)related to photosynthetic pigment.We can get up to 27 pathway branches of porphyrin and chlorophyll metabolism pathway(ko00860),and 17 pathway branches of carotenoid biosynthesis pathway(ko00906).66 differential expression gene(DEGs)related to photosynthetic pigment were screened out,28 of them were up regulated and 38 were down regulated.The selected photosynthetic pigment related pathways,DEGs,were predicted.28 structural genes were screened out in 66 DEGs,and 19 key enzymes(POR,DVR,NOL,LCYB,NCED,CYP707A)were selected from 66 DEGs,and they can be divided into four types:chlorophyll biosynthesis,chlorophyll degradation,carotenoid synthesis and carotenoid degradation.The screened structural genes were compared with the protein sequences in the KOG database and the GO function classification statistics were carried out.The most abundant ones were biological process,among which the most significant functional enrichment was related to metabolism process,single biological process and cell process.The screening of photosynthetic pigment related genes will help to reveal the molecular mechanism of leaf color formation ofAcer pseudosieboldianum,and provide data support for the future study on gene localization and gene cloning of Acer pseudosieboldianum.
