摘要
MADS-box基因家族与植物生长和发育调控密切相关。为揭示油莎草MADS-box家族成员组织表达模式,本研究根据油莎草茎块RNA-Seq结果,利用HMM搜索鉴定了26个家族成员,系统发育分析及保守结构域分析表明油莎草MADS-box成员主要属于MIKC型,具有典型的MEF2类MADS结构域,在不同茎块发育时期具有7种表达模式,其中CeMADSXAL1基因在茎块发育初期高表达,推测CeMADSXAL1基因对油莎草茎块发育过程具有重要调控作用。此外,研究对CeMADSXAL1基因进行克隆,获得长度为747 bp的开放阅读框,编码249个氨基酸;亚细胞定位分析结果显示,Ce MADSXAL1蛋白主要在细胞核内表达,结果为进一步阐明油莎草MADS-box基因功能提供了理论基础。
The MADS-box gene family is intimately related to growth and regulation of development in plant.In the light of the results of RNA-Seq for tuber of Cyperus esculentus,the results of this research were as follows.To proclaim the expression pattern of MADS-box family members.26 family members were identified by HMM search,phylogenetic and conserved domain analysis showed that MADS-box members of Cyperus esculentus be-longed to MIKC type,which has a typical MEF2 like domain.There were 7 expression patterns in different tuber development stages,CeMADSXAL1 gene was highly expressed at the early stage of tuber development.It can be inferred that CeMADSXAL1 gene plays an extraordinary important role in the development of Cyperus esculentus tuber.In the meantime,the CeMADSXAL1 gene was cloned and an open reading frame of 747 bp was obtained,which encoded 249 amino acids,CeMADSXAL1 was found strongly signal in nucleus,that result of the subcellular localization analysis.This study was concluded the theoretical basis for further elucidating the function of MADS-box gene in Cyperus esculentus.
