铁死亡在肿瘤诊断和治疗中的研究进展

摘要 铁死亡是一种有别于自噬、凋亡、坏死和焦亡的新型程序性细胞死亡方式,其特点是细胞内过多的铁离子蓄积,导致脂质过氧化反应,最终引发细胞膜破裂和细胞死亡。这种独特的细胞死亡模式受多种细胞内代谢事件包括氧化应激、铁代谢、线粒体活动、氨基酸、脂质和糖的代谢等调节。近年来研究表明,铁死亡参与到肿瘤发生、发展等多个生物学过程当中,提示铁死亡在肿瘤诊断和治疗当中具有巨大的潜力。诱导铁死亡可以有效抑制肿瘤的生长和转移,同时增加肿瘤细胞对其他药物的治疗敏感性。该文系统总结了铁死亡相关的分子调控机制,以及它在肿瘤诊断和治疗当中的潜在应用价值。 Ferroptosis is a novel way of regulated cell death,which is different from other forms of cell death,such as autophagy,apoptosis,necrosis and pyrodeath.It is characterized by excessive accumulation of lipid peroxides in an iron-dependent manner,resulting in cell membrane rupture and cell death.This unique pattern of cell death is regulated by a variety of intracellular metabolic events,including oxidative stress,iron handling,mitochondrial activity,and metabolism of amino acids,lipids and sugars.Recent studies have shown that ferroptosis is involved in biological process of tumor growth and progression,suggesting that ferroptosis has a great potential in tumor diagnosis and treatment.Ferroptosis inducers could effectively inhibit tumor growth and metastasis,while contribute the therapeutic sensitivity of tumor cells.This review systematically summarizes the molecular regulatory mechanism underlying ferroptosis,and provides its potential application in tumor diagnosis and treatment.
