CeO_(2)纳米酶对环磷酰胺所致斑秃小鼠毛发生长的影响

摘要 目的 探讨氧化铈(CeO_(2))纳米酶对环磷酰胺所致斑秃小鼠模型毛发生长的影响。方法 将雌性C57BL/6J小鼠30只随机分为空白组、模型组和CeO_(2)纳米酶组(300μg/mL),每组10只。模型组和CeO_(2)纳米酶组小鼠采用单次腹腔内注射环磷酰胺注射液,构建斑秃小鼠模型。CeO_(2)纳米酶组小鼠给予300μg/mL CeO_(2)纳米酶外涂于小鼠背部剃毛区,空白组和模型组小鼠均予以等体积的PBS溶液外涂,1次/d,连续27 d。实验结束后(第28天)肉眼观察小鼠背部秃发区毛发生长情况并进行评分;HE染色小鼠背部经处理的皮肤组织计算单位视野内毛囊数量;酶联免疫吸附试验(ELISA)法检测小鼠血清细胞间黏附分子-1(ICAM-1)及内皮细胞白细胞黏附分子-1(ELAM-1)水平;水溶性四唑盐-1法(WST-1)检测皮肤组织丙二醛(MDA)及超氧化物歧化酶(SOD)含量。结果 与空白组相比,模型组小鼠毛发生长及毛发评分显著下降(P<0.05),毛囊数量明显减少(P<0.05),血清ICAM-1及ELAM-1水平升高(P<0.05),皮肤组织MDA含量升高而SOD含量降低(P<0.05);与模型组相比,CeO_(2)纳米酶组毛发生长及毛发评分显著增加(P<0.05),毛囊数量明显增多(P<0.05),血清ICAM-1及ELAM-1水平降低(P<0.05),皮肤组织MDA含量降低而SOD含量升高(P<0.05)。结论 外用CeO_(2)纳米酶对环磷酰胺所致斑秃小鼠模型具有促进毛发再生的作用,为CeO_(2)纳米酶外用治疗斑秃提供了实验依据。 Objective To investigate the effect of CeO_(2) nanoenzyme on the hair growth of cyclophosphamide-induced alopecia areata mice.Methods Thirty female C57BL/6J mice were randomly divided into blank group,model group and CeO_(2) nanoenzyme(300μg/mL)group,with 10 mice in each group.Mice in model group and CeO_(2) nanoenzyme group were intraperitoneally injected with cyclophosphamide once to establish the model of alopecia areata.Mice in CeO_(2) nanoenzyme group were coated with 300μg/mL CeO_(2) nanoenzyme on the back shaving area of mice,and mice in blank control group and model group were coated with equal volume of PBS solution once a day for 27 d.After the experiment(at day 28),the hair growth in the bald area of the back of mice was observed and scored.The number of hair follicles per unit visual field was calculated in the treated back skin tissue of mice stained by HE.Serum levels of intercellular adhesion molecule-1(ICAM-1)and endothelium-leukocyte adhesion molecule-1(ELAM-1)were determined by enzyme-linked immunosorbent assay(ELISA).The contents of malondialdehyde(MDA)and superoxide dismutase(SOD)in skin tissue were determined by water-soluble tetrazole salt-1 method(WST-1).Results Compared with blank group,hair growth and hair score were significantly decreased in model group(P<0.05),the number of hair follicles was significantly decreased(P<0.05),serum ICAM-1 and ELAM-1 levels were increased(P<0.05),MDA content in skin tissue was increased(P<0.05),and SOD content was decreased(P<0.05).Compared with model group,hair growth and hair score were significantly increased in CeO_(2) nanoenzyme group(P<0.05),the number of hair follicles was significantly increased(P<0.05),serum ICAM-1 and ELAM-1 levels were decreased(P<0.05),MDA content in skin tissue was decreased(P<0.05),and SOD content was increased(P<0.05).Conclusion Topical CeO_(2) nanoenzymes has therapeutic effect on cyclophosphamide-induced alopecia areata of mice,which provides a theoretical basis for the external application of CeO_(2) nanoenzymes in the treatment of alopecia areata in the future.
