基于化学激活荧光素酶表达基因法和标准方法评估动物源性固态食品中17种二噁英类化合物毒性当量的相关性比较

摘要 为使化学激活荧光素酶表达基因法(CALUX)在动物源性固态食品中二噁英类化合物毒性当量筛查中具有更好的适用性,使其能够与标准方法高分辨气相色谱-高分辨质谱法(HRGC-HRGC)的评估结果相互转换,分别基于CALUX和HRGC-HRGC评估了肉类、海鲜类和配方乳粉这3类典型动物源性固态食品中7种2,3,7,8-取代的多氯代二苯并二噁英(PCDDs)和10种2,3,7,8-取代的多氯代二苯并呋喃(PCDFs)的毒性当量,分析两种方法测定结果的相关性,并确定换算关系和适宜样品类型。结果表明:基于HRGC-HRMS得到的样品中17种目标物的毒性当量(TEQ)下限值为0.001~0.106 pg·g^(-1),TEQ上限值为0.004~0.242 pg·g^(-1),远低于欧盟法规EC 1881/2006规定的限量值(3.5 pg·g^(-1));基于CALUX得到的肉类样品中17种目标物的毒性当量(BEQ)值为0.74~1.30 pg·g^(-1),海鲜类样品中17种目标物的BEQ值为0.31~0.63 pg·g^(-1),配方乳粉样品中17种目标物的BEQ值为0.043~0.074 pg·g^(-1);肉类和配方乳粉样品的BEQ值与TEQ值的相关性较差,而海鲜类样品的BEQ值与TEQ上限值相关性较好。 In order to improve the applicability of chemically activated luciferase expression gene method(CALUX)in screening for toxic equivalents of dioxins in solid foods of animal origin,and to make it interconvertible with the evaluation results of the standard method high resolution gas chromatography-high resolution mass spectrometry(HRGC-HRGC),the toxic equivalents of 7 kinds of 2,3,7,8-substituted polychlorinated dibenzo-dioxins(PCDDs)and 10 kinds of 2,3,7,8-substituted polychlorinated dibenzo-furans(PCDFs)in three kinds of typical solid foods of animal origin including meat,seafood and formula milk based on CALUX and HRGC-HRGC were evaluated,and the correlation between the determined results of two methods was analyzed,and the conversion relationship and suitable sample type were determined.It was shown that the toxic equivalent(TEQ)lower limits of 17 targets in the samples based on HRGC-HRMS ranged from 0.001 to 0.106 pg·g^(-1),and the TEQ upper limits ranged from 0.004 to 0.242 pg·g^(-1),that were much lower than the limits of 3.5 pg·g^(-1)stipulated in European Union regulation EC 1881/2006.The toxic equivalent(BEQ)values of 17 targets in meat,seafood,and formula milk powder samples based on CALUX were in the ranges of 0.74-1.30,0.31-0.63 pg·g^(-1),and 0.043-0.074 pg·g^(-1),respectively.The correlation between BEQ and TEQ values of meat and formula milk samples was poor,while the correlation between BEQ and TEQ upper limit values of seafood samples was good.
