Rab35、TRIM8在子宫内膜癌中的表达及其与患者临床病理特征和预后的关系

Expression of Rab35 and TRIM8 in endometrial cancer and their relationship with clinicopathological features and prognosis of patients

目的探讨Rab35、三结构域蛋白8(TRIM8)在子宫内膜癌(EC)中的表达,并分析其与患者临床病理特征和预后的关系。方法选择2017年1月至2020年1月沧州市人民医院收治的180例EC患者,取手术切除的癌组织和癌旁组织。采用免疫组化法检测Rab35、TRIM8表达,比较EC癌组织和癌旁组织中Rab35、TRIM8表达差异,以及不同临床病理特征患者EC组织中Rab35、TRIM8表达阳性率差异。采用多因素Logistic回归分析影响EC患者预后的因素,采用Kaplan-Meier生存曲线分析EC患者生存情况。结果EC癌组织Rab35表达阳性率高于癌旁组织(P<0.05),TRIM8表达阳性率低于癌旁组织(P<0.05)。FIGO分期ⅢA期、低分化、肌层浸润深度≥1/2、有淋巴结转移的EC患者癌组织Rab35表达阳性率高于FIGO分期Ⅰ~Ⅱ期、中高分化、肌层浸润深度<1/2、无淋巴结转移的EC患者(P<0.05),FIGO分期ⅢA期、低分化、肌层浸润深度≥1/2、有淋巴结转移的EC患者癌组织TRIM8表达阳性率低于FIGO分期Ⅰ~Ⅱ期、中高分化、肌层浸润深度<1/2、无淋巴结转移的EC患者(P<0.05)。多因素Logistic回归分析结果显示FIGO分期ⅢA期、有淋巴结转移、Rab35表达阳性是EC患者死亡的危险因素(P<0.05),TRIM8表达阳性是EC患者死亡的保护因素(P<0.05)。Rab35表达阳性EC患者的累积生存率低于Rab35表达阴性患者(Log-Rankχ~2=11.523,P<0.001);TRIM8表达阳性EC患者的累积生存率高于TRIM8表达阴性患者(Log-Rankχ~2=9.683,P<0.001)。结论EC组织中Rab35表达阳性率上调,TRIM8表达阳性率下调,且与EC低分化、FIGO分期ⅢA期、肌层浸润深度、淋巴结转移以及低生存率有关。

Objective To investigate the expressions of Rab35 and tridomain protein 8(TRIM8) in endometrial cancer(EC),and to analyze their relationship with clinicopathological features and prognosis of the patients.Methods A total 180 patients with EC admitted and treated in this hospital from January 2017 to January 2020 were selected,and the surgically resected cancer tissues and paracancerous tissues were taken.Immunohistochemicall method was employed to detect the expression of Rab35 and TRIM8,the differences in positive expressions of Rab35 and TRIM8 between cancer tissure and paracancerous tissue were compared,as well as the differences in positive expressions of Rab35 and TRIM8 in cancer tissure among the EC patients with different clinicopathological features.The multivariate Logistic regression was adopted to analyze the factors affecting the prognosis of the EC patients,and the Kaplan-Meier survival curve was adopted to analyze the survival situation of the patients with EC.Results The expression positive rate of Rab35 in EC cancer tissues was higher than that in paracancerous tissues(P<0.05),and the expression positive rate of TRIM8 was lower than that in paracancerous tissues(P<0.05).The expression positive rate of Rab35 in EC cancer tissues of FIGO stage ⅢA,low differentiation,depth of myometrial invasion ≥1/2 and lymph node metastasis was higher than that in EC cancer tissues of FIGO stage Ⅰ—Ⅱ,medium-high differentiation,depth of myometrial invasion <1/2 and non-lymph node metastasis(P<0.05),the expression positive rate of TRIM8 in EC cancer tissues of FIGO stage ⅢA,low differentiation,depth of myometrial invasion ≥1/2 and lymph node metastasis was lower than that in EC cancer tissues of FIGO stage Ⅰ—Ⅱ,medium-high differentiation,depth of myometrial invasion <1/2 and non-lymph node metastasis(P<0.05).Multivariate Logistic regression analysis showed that FIGO stage ⅢA,lymph node metastasis and Rab35 expression positive were the risk factors for the mortality in EC patients(P<0.05),and TRIM8 expression positive was a protective factor for mortality in EC patients(P<0.05).The cumulative survival rate in EC patients with Rab35 expression positive was lower than that in EC patients with Rab35 expression negative(Log-Rank χ~2=11.523,P<0.001);the cumulative survival rate in EC patients with TRIM8 expression positive was higher than that in the EC patient with TRIM8 expression negative(Log-Rank χ~2=9.683,P<0.001).Conclusion The expression positive rate of Rab35 in EC tissues is up-regulated,and the expression positive rate of TRIM8 is down-regulated,moreover which is related to the low differentiation of EC,FIGO stage ⅢA,depth of myometrial invasion,lymph node metastasis and low survival rate.
