摘要
目的 采用生物信息学方法探讨COL1A2在头颈部鳞癌(HNSC)中的表达及相关功能。方法 利用TIMER2.0数据库分析COL1A2基因在泛癌中的表达;通过GEPIA数据库、UALCAN数据库分析该基因在HNSC与正常组织中的差异表达情况;并利用STRING数据库构建COL1A2基因蛋白互作网,DAVID数据库分析与该基因互作蛋白基因的相关功能;最后通过GCBI基因雷达数据库预测该基因可能参与的调控网络和转录因子。结果 COL1A2在多种肿瘤中高表达,且在头颈部鳞癌组织与正常组织中有差异表达。COL1A2及相关蛋白基因功能富集分析发现其参与了4个细胞组分、5个分子功能、11个生物学程序、7条信号通路,在基因调控网及转录因子预测发现,COL1A2与多种蛋白、转录因子、lncRNA、micRNA等相互调控。结论 COL1A2与HNSC的发生、发展可能是通过PI3K-Akt信号通路等多种途径、多种蛋白、转录因子共同调控的结果。
Objective To investigate the expression and related functions of COL1A2 in head and neck squamous cell carcinoma(HNSC).Methods The expression of COL1A2 gene in generalized carcinoma was analyzed by using TIMER 2.0 database.GEPIA database and UALCAN database were used to analyze the differential expression of this gene in HNSC and normal tissues.The protein interaction network of the COL1A2 gene was constructed by the STRING database,and the related functions of the gene interaction protein gene were analyzed by DAVID database.Finally,GCBI gene radar database was used to predict the regulatory network and transcription factors that the gene might be involved in.Results COL1A2 was highly expressed in various tumors,and was differentially expressed in head and neck squamous cell carcinoma and normal tissues.Functional enrichment analysis of COL1A2 and related protein genes showed that COL1A2 was involved in 4 cell components,5 molecular functions,11 biological programs and 7 signaling pathways.In gene regulatory network and transcription factor prediction,COL1A2 was considered to be mutually regulated with a variety of proteins,transcription factors,lncRNA,and micRNA,etc.Conclusion The occurrence and development of COL1A2 with HNSC may be the result of the joint regulation of various pathways,various proteins,and transcription factors through the PI3K-Akt signaling pathway.
