微管蛋白α-1B在泛癌中的表达及临床意义

摘要 目的利用生物信息学分析系统阐述微管蛋白α-1B(TUBA1B)与泛癌特征的关系。方法利用TCGA数据库中的肿瘤患者临床资料,从表达水平、疾病分期、预后、基因突变特征、免疫浸润水平和基因富集分析6个方面探讨TUBA1B与各种肿瘤类型的相关性。结果TUBA1B的表达水平在多数肿瘤中高于癌旁(P<0.05),在包括侵袭性乳腺癌等14种肿瘤中具有早期诊断价值(P<0.05),在肾上腺皮质癌等3种肾脏肿瘤中具有晚期进展监测价值(P<0.05),且高表达TUBA1B在多种肿瘤预示较差预后(P<0.05)。免疫浸润分析提示TUBA1B与泛癌中多数免疫浸润细胞及多种免疫相关基因表达呈正相关(P<0.05)。基因富集分析表明TUBA1B可能通过调控细胞周期、DNA损伤修复、有丝分裂等生物学过程参与肿瘤发生发展。结论TUBA1B参与了多种肿瘤的发生发展和免疫浸润,可作为泛癌的肿瘤标志物和潜在的药物靶点。 Objective To illustrate the relationship between tubulin alpha-1B(TUBA1B)and pan-cancer features through bioinformatics analysis.Methods Clinical data of cancer patients in TCGA database were used to explore the correlation between TUBA1B and various cancer types from six aspects:expression levels,disease staging,prognosis,gene mutation characteristics,immune infiltration level and gene enrichment analysis.Results TUBA1B was highly expressed in most cancers compared with adjacent non-tumor tissues(P<0.05).TUBA1B showed early diagnostic value in 14 tumors,including invasive breast cancer(P<0.05),and late progression monitoring value in 3 renal tumors,including adrenocortical carcinoma(P<0.05).High expression of TUBA1B indicated a poor prognosis in multiple tumors(P<0.05).Immunoinfiltration analysis revealed that TUBA1B was positively correlated with most immune-infiltrating cells and immune-related genes in pan-cancer(P<0.05).Gene enrichment analysis indicated that TUBA1B may be involved in the development of tumor by regulating biological processes such as cell cycle,DNA damage repair,and mitosis.Conclusion TUBA1B is involved in the occurrence,development and immunoinfiltration of a variety of tumors,which can be used as a tumor marker for pan-cancer and a potential drug target.
