摘要
目的应用动物模型对比两种愈合方式促进寒区放射性皮肤损伤修复效果。方法30只SD大鼠随机分为对照组、37℃水凝胶组和红外线照射组,每组10只,在-20℃环境下饲养1周,采用锶90建立大鼠寒区放射性损伤模型。37℃水凝胶组和红外线照射组分别于每日采取37℃水凝胶覆盖伤口和红外线照射后无菌纱布覆盖伤口,记录大鼠创口愈合时间、创面愈合率;HE染色观察皮肤病理改变;ELISA检测血清炎症因子IL-1β、IL-6及TNF-α表达水平;Western blot检测Ki67和Cytokeratin 19(CK19)蛋白表达。结果与对照组和红外线组相比,37℃水凝胶组大鼠放射性皮肤损伤愈合时间最短,创面愈合率最高,换药31天时皮肤愈合情况显著优于其他两组;在换药第10天,37℃水凝胶组皮肤损伤减轻明显,上皮恢复较其他两组明显;37℃水凝胶组大鼠创面组织中IL-1β、IL-6及TNF-α表达水平最低;37℃凝胶干预后Ki67和CK19蛋白表达水平最高。结论37℃水凝胶相比红外线照射更有效促进寒区放射性皮肤损伤愈合,且使用方便,适于临床推广。
Objective Comparing the effect of two different therapy methods repairing radiation-induced skin injury in cold region of contrast agent in animal models.Methods Thirty SD rats were randomly divided into model group,37℃hydrogel group and infrared irradiation group,with 10 rats in each group.The rats were fed at-20℃for 1 week,and the radiation damage model of rats in cold area was established by using strontium 90.The rats of 37℃hydrogel group and infrared irradiation group were respectively covered with 37℃hydrogel and sterile gauze after infrared irradiation daily.The wound healing time and wound healing rate of rats were recorded.The pathological changes of skin was observed by HE staining.The expression levels of IL-1β,IL-6 and TNF-αin serum were detected by ELISA.The expressions of Ki67 and Cytokeratin 19(CK19)were detected by Western blot.Results Compared with control group and infrared group,the healing time of radioactive skin injury in 37℃hydrogel group was the shortest and the wound healing rate was the highest,and the skin healing rate at 31 days after dressing change was significantly better than the other two groups.On the 10th day of dressing change,the skin injury of 37℃hydrogel group was significantly reduced,and the epithelial recovery was more obvious than that of the other two groups.The expressions of IL-1β,IL-6 and TNF-αwere the lowest in the 37℃hydrogel group.The expression levels of Ki67 and CK19 protein were the highest after 37℃gel intervention.Conclusion Compared with infrared irradiation,gel at 37℃was more effective and beneficial to skin healing in cold regions,and it is convenient to use and suitable for clinical promotion.
