摘要
目的 研究人参总皂苷对博莱霉素诱导的肺纤维化小鼠肺组织中促炎细胞因子IL-6和IL-1β的影响。方法 随机将60只雄性昆明种小鼠分为正常对照组、模型组、以及阳性药吡菲尼酮组(100 mg/kg)和人参总皂苷(40、80、160 mg/kg)组;除正常对照组外,其余5组均通过气管单次滴注博莱霉素(3 mg/kg)构建小鼠肺纤维化模型。建模次日灌胃给药,每天1次,连续给药14 d,取肺组织观察其病理形态、肺部炎症和纤维化改变程度。检测肺纤维化小鼠肺组织中Ⅰ型胶原蛋白(COLⅠ)、白细胞介素-6(IL-6)以及白细胞介素-1β(IL-1β)蛋白水平。结果 气管内单次滴注博莱霉素14 d后,模型组小鼠肺组织明显淤血实变,肺泡炎及肺纤维化明显,肺组织中COLⅠ、IL-6及IL-1β蛋白水平较正常对照组显著上调。与模型组相比,人参总皂苷及吡菲尼酮能够有效改善模型小鼠肺泡炎及肺纤维化程度,且明显减少肺组织中COLⅠ、IL-6及IL-1β蛋白水平。结论 人参总皂苷改善博莱霉素诱导的小鼠肺纤维化与抑制炎症因子IL-6及IL-1β蛋白水平有关。
Objective To study the effects of total ginsenosides on pro-inflammatory cytokines IL-6 and IL-1βin lung tissues of mice with bleomycin-induced pulmonary fibrosis.Methods Sixty male Kunming mice were randomly divided into a normal control group,a model group,and a positive drug pirfenidone group(100 mg/kg)and total ginsenosides(40,80,160 mg/kg)groups;Except for the normal control group,the remaining five groups were constructed with a mouse lung fibrosis model by single-dose drops of bleomycin(3 mg/kg)through the oral endotracheal tube.The drug was administered by gavage on the next day of modelling,once a day for 14 consecutive days,and the lung tissues were taken to observe the pathological morphology,the degree of inflammation and fibrotic changes in the lungs.Type-I collagen(COLⅠ),interleukin-6(IL-6)and interleukin-1β(IL-1β)protein levels were measured in the lung tissues of mice with pulmonary fibrosis.Results After 14 days of intraoral tracheal drip injection of bleomycin,the lung tissues of mice in the model group were obviously bruised and solid,alveolitis and pulmonary fibrosis were obvious,and the levels of COLⅠ,IL-6 and IL-1βprotein in the lung tissues were increased significantly in comparison with those in the normal control group.When compared with the model group,total ginsenosides and pirfenidone effectively improved the degree of alveolitis and pulmonary fibrosis in the model mice,and significantly reduced the levels of COLⅠ,IL-6 and IL-1βprotein in the lung tissues.Conclusion Total ginsenosides ameliorated bleomycin-induced pulmonary fibrosis in mice in association with the inhibition of inflammatory factors IL-6 and IL-1βprotein levels.
