氯化-N-十二烷基吡啶-1-乙酰胺杀灭或抑制病原体的活性研究

摘要 季铵盐类表面活性剂是一类低毒、高效的广谱消毒剂,能杀灭多种细菌和病毒,在养殖业和医学等领域的消毒中应用前景广阔。研究旨在明确自主合成的季铵盐类表面活性剂氯化-N-十二烷基吡啶-1-乙酰胺(C12cmpC)对禽源致病菌和牛病毒性腹泻病毒(BVDV)的抗菌抗病毒活性。通过MIC、MBC和时间杀菌试验,明确C12cmpC的体外杀灭、抑制革兰阴性和阳性细菌的效果。通过细胞形态观察、细胞活性检测和BVDV m RNA的qRT-PCR检测,初步明确C12cmpC的抗病毒活性。结果表明,C12cmpC对鹅大肠杆菌(QS195313)的MIC为250 mg·L^(-1),MBC为500 mg·L^(-1)。C12cmpC对大肠杆菌和金黄色葡萄球菌标准株的MIC为31.25 mg·L^(-1),MBC为62.5 mg·L^(-1)。此外,C12cmpC对MDBK细胞的最大安全浓度为5 mg·L^(-1)。其与BVDV作用1、5、30、60 min后,MDBK细胞中病毒m RNA水平均显著降低。研究表明新型季铵盐类表面活性剂氯化-N-十二烷基吡啶-1-乙酰胺具有较好的抗菌抗病毒活性,研究为新型消毒剂的开发提供了理论依据。 Quaternary ammonium surfactants are a kind of low toxicity and high efficiency broad-spectrum disinfectants,which can kill a variety of bacteria and viruses,and have broad application prospects in the disinfection of breeding industry and medical fields.To study the antibacterial and antiviral activity of N-dodecyl-2-(piridin-1-ium)acetamid(C12cmpC),a self-synthesized quaternary ammonium surfactant,against avian pathogens and bovine viral diarrhea virus(BVDV).The efficacy of C12cmpC in killing or inhibiting gram-negative and gram-positive bacteria in vitro was determined by MIC,MBC and time bactericidal tests.The antiviral activity of C12cmpC was preliminarily determined by cell morphology observation,cell activity detection and qRT-PCR detection of BVDV mRNA.The results showed that the MIC and MBC of C12cmpC against Escherichia coli(QS195313)were 250 mg·L^(-1) and 500 mg·L^(-1) respectively.The MIC and MBC of C12cmpC against Escherichia coli and Staphylococcus aureus were 31.25 mg·L^(-1) and 62.5 mg·L^(-1) respectively.In addition,the maximum safe concentration of C12cmpC for MDBK cells was 5 mg·L^(-1).The viral mRNA levels in MDBK cells were significantly decreased after 1,5,30 and 60 min treatment with BVDV.The results show that the new quaternary ammonium surfactant N-dodecyl-2-(piridin-1-ium)acetamide has good antibacterial and antiviral activity.This study provides a theoretical basis for the development of new disinfectants.
