基于功能化介孔二氧化硅的贻贝中磷脂选择性纯化研究

摘要 为制备高纯度海洋源磷脂,本研究利用微乳液法制备介孔二氧化硅(KCC-1)微球,并基于水蒸气内部水解法对其进行功能化修饰,分别合成二氧化钛/KCC-1(TiO_(2)/KCC-1)和氧化石墨烯/KCC-1(GO/KCC-1)复合材料,再通过扫描电子显微镜(SEM)、透射电子显微镜(TEM)、原子力显微镜(AFM)等技术表征形貌结构,而后制成固相微萃取(SPE)柱。以贻贝为原料,分别采用TiO_(2)/KCC-1 SPE和GO/KCC-1 SPE法纯化磷脂,以富集因子(EF)作为指标评价其性能,达到制备高纯度磷脂的目的。结果表明,合成的KCC-1微球直径为250~400 nm,形貌良好、大小均一、比表面积较大,且经功能化修饰,成功合成了TiO_(2)/KCC-1和GO/KCC-1填料。对比分析可知,EF_(TiO_(2)/KCC-1)>EF_(GO/KCC-1),说明TiO_(2)/KCC-1 SPE法纯化磷脂的作用效果更优。通过该方法制备的磷脂纯度高达93.57%。本研究结果为高纯度海洋源磷脂的制备提供了技术支撑。 To prepare high purity marine source phospholipids,the functionalized mesoporous core-shell(KCC-1)microspheres were prepared by the microemulsion method.Furthermore,the titanium dioxide/KCC-1(TiO_(2)/KCC-1)and graphene oxide/KCC-1(GO/KCC-1)composites were prepared by the water vapor internal hydrolysis.The morphology and structure of the prepared materials were characterized by scanning electron microscopy(SEM),transmission electron microscopy(TEM)and atomic force microscopy(AFM).The two new materials were used as the adsorbent to prepare solid phase extraction(SPE)column.The phospholipids extracted from mussels were purified by TiO_(2)/KCC-1 SPE and GO/KCC-1 SPE,respectively.The performance of these two methods was evaluated by the enrichment factor(EF).Finally,the high purify phospholipids were successfully prepared by the superior SPE method.As a result,the microspheres of synthesized KCC-1 were 250-400 nm in diameter with good morphology,uniformed size and large specific surface area.After functionalized modification,the TiO_(2)/KCC-1 and GO/KCC-1 composites were successfully prepared.It was found that the EF_(TiO_(2)/KCC-1)was greater than EFGO/KCC-1,indicating that TiO_(2)/KCC-1 SPE method had a superior effect on the purification of phospholipids.The phospholipid prepared by TiO_(2)/KCC-1 SPE method had a purity of 93.57%.The results of this study could provide more technical support for the preparation of high purity marine source phospholipids.
