茉莉酸甲酯和酵母提取物对白鲜试管无菌苗生长和药用成分含量的影响 被引量:1

Effect of Methyl Jasmonate and Yeast Extract on the Growth and Medicinal Component Content of Dictamnus Test-Tube Aseptic Seedlings

摘要 为探究最适白鲜幼苗生长以及提高其药用成分含量的茉莉酸甲酯(MeJA)和酵母提取物(YE)浓度,以白鲜种子为试验材料,以不加诱导子的MS培养基作为对照处理(CK),观察白鲜幼苗在不同MeJA和YE添加浓度的MS培养基中的生长情况,并采用高效液相色谱法(HPLC)测定白鲜碱、梣酮和黄柏酮含量。结果表明,不同浓度MeJA和YE能影响白鲜幼苗的生长以及药用成分含量。其中,白鲜幼苗根长、侧根数、生物量和三种药用成分含量均随MeJA浓度的增加呈现先上升后下降趋势,在1μmol·L^(-1)时,白鲜幼苗根长、侧根数、鲜重净增量和干重净增量达到最大值,分别是CK的1.78、1.51、2.38和1.28倍,5μmol·L^(-1)时白鲜碱、黄柏酮含量最高,分别是CK的2.88和3.13倍,0.1μmol·L^(-1)时梣酮含量提升最为明显,是CK的3.64倍。对YE而言,白鲜幼苗根长、侧根数、生物量和三种药用成分含量均随YE浓度的增加呈现先上升后下降趋势,在30 mg·L^(-1)时对白鲜幼苗的根长、侧根数、鲜重净增量和干重净增量提升最为明显,分别是CK的2.40、3.23、2.62和1.27倍,10 mg·L^(-1)处理后白鲜碱含量最高,是CK的2.74倍,100 mg·L^(-1)时梣酮含量提升最为明显,是CK的2.12倍,30 mg·L^(-1)时黄柏酮含量最高,是CK的2.77倍。主成分分析结果显示,MeJA的最适诱导浓度为1μmol·L^(-1),YE最适诱导浓度为30 mg·L^(-1),以30 mg·L^(-1)YE效果最佳。本研究为提升药用植物白鲜中次生代谢产物的含量提供了参考。 The aim of this study was to explore the optimal concentration of methyl jasmonate(MeJA)and yeast extract(YE)on the growth of Dictamnus seedlings,as well as their role to increase the content of its medicinal components.Thus,the seeds of Dictamnus were used as experimental materials.Our approach was to observe the growth of Dictamnus seedlings in MS-medium supplemented with different concentrations of MeJA and YE,and the MS-medium without addition of elicitors was used as control(CK).Contents of dictamnine,fraxinellone and obacunone were determined by high performance liquid chromatography(HPLC).Results showed that MeJA and YE greatly affected the growth of Dictamnus seedlings and the content of medicinal components.The content of root length,lateral root number,biomass and three medicinal components of Dictamnus seedlings increased first and then decreased with the increase of MeJA concentration.The root length,lateral root number,fresh weight increment and dry weight increment of Dictamnus seedlings reached the maximum at 1µmol·L^(-1)of MeJA,which were 1.78,1.51,2.38 and 1.28 times of the control treatment(CK),respectively.The contents of dictamnine and obacunone were the highest at 5µmol·L^(-1)of MeJA,which were 2.88 and 3.13 times of CK.The content of fraxinellone increased the most at 0.1µmol·L^(-1)of MeJA,which was 3.64 times of CK.Similarly,the content of root length,lateral root number,biomass and three medicinal components of Dictamnus seedlings increased first and then decreased with the increase of YE concentration.At 30 mg·L^(-1)of YE,the root length,lateral root number,net increment of fresh weight and net increment of dry weight of Dictamnus seedlings increased the most,which were 2.40,3.23,2.62 and 1.27 times of CK,respectively.The content of dictamnine was the highest after 10 mg·L^(-1)treatment by YE,which was 2.74 times of CK.The content of fraxinellone increased the most at 100 mg·L^(-1)of YE,which was 2.12 times of CK.The content of obacunone was the highest at 30 mg·L^(-1)of YE,which was 2.77 times that of CK.Based on the principal component analysis,the optimal induction concentration of MeJA was 1µmol·L^(-1),and the optimal induction concentration of YE was 30 mg·L^(-1).Overall,this research provides more information to improve the content of secondary metabolites in medicinal plant Dictamnus.
