土培和水培条件下两种党参有效成分含量的比较

Content comparison of effective components for two kinds of Codonopsis pilosula cultivated in soil and hydroponics conditions

摘要 采用反相高效液相色谱(RP-HPLC)法测定了室内土培和水培条件下渭党和纹党中紫丁香苷、党参炔苷和苍术内酯Ⅲ3种活性成分的含量,以期比较室内不同栽养方式对2种党参有效成分积累水平的影响.色谱流动相采用乙腈(A)-0.3%磷酸水溶液(B),体积流量为1 mL·min^(-1),检测波长为220 nm,柱温为30℃.结果表明,3种受测成分在各自质量浓度范围内线性关系良好(R 2>0.9998),土培纹党中紫丁香苷、苍术内酯Ⅲ含量较高,分别为0.025,0.020 mg·g^(-1);水培渭党中党参炔苷含量较高,为3.944 mg·g^(-1).实验重复性和稳定性良好,精密度和准确性可靠.2种栽养条件下,纹党和渭党中3种活性成分含量差异显著,研究结果可为党参栽培方式的选择和优良品种的筛选提供数据参考. RP-HPLC was applied to measure the quantity of syringin,lobetyolin and atractylenolideⅢin Codonopsis pilosula Nannf.var.Modesta(Cpm)and C.pilosula(Cp)under two conditions of soil culture and hydroculture to analyze the three components differences of the two taxa between two cultivation methods.The chromatography system included that mobile phase is acetonitrile(A)-0.3%phosphoric acid aqueous solution(B),the flow rate is 1 mL·min^(-1),the detection wavelength is 220 nm,and the column temperature is 30℃.The results showed that the linear relationship between the three components is quite well(R 2>0.9998),and the contents of syringin and atractylenolideⅢin soil culture of Cpm are higher than others,which is 0.025 and 0.020 mg·g^(-1),respectively.The higher content of lobetyolin is 3.944 mg·g^(-1) in hydroponic Cp.The experiment has good repeatability and stability,as well as reliable precision and accuracy.The contents of three active ingredients in Cpm and Cp are significantly different under the two cultivation conditions.The results may provide data reference for the selection of cultivation methods and selective breeding of excellent varieties.
