当归痛风方对痛风性关节炎大鼠滑膜的傅里叶变换红外光谱仪研究

摘要 目的:通过傅里叶变换红外光谱仪(FTIR)观察当归痛风方对痛风性关节炎(GA)大鼠滑膜的干预效果,为动物疾病模型判定和药物评价提供新的途径。方法:60只SD大鼠随机分为6组:正常对照组、模型组、阳性药组和当归痛风方低、中、高剂量组,采用腹腔注射氧嗪酸钾和膝关节腔尿酸钠的方法复制GA模型,造模1周后,灌胃当归痛风方13.08、26.16、52.32 g·kg^(-1)·d^(-1),检测或观察大鼠血清尿酸(UA)及关节液IL-6、IL-1β含量,病理切片,FTIR谱图及其二阶导数谱图。结果:与模型组比较,当归痛风方高剂量组大鼠体质量第21天显著增长(P<0.05),血清UA及关节液IL-6、IL-1β含量显著下降(P<0.05),并改善了滑膜的病理变化,FTIR二阶导数谱图接近正常对照组。结论:FTIR检测方法一定程度上可作为评价当归痛风方改善GA疗效的检测手段之一,对GA动物模型判定及药物评价具有重要的参考意义,并拓展了FTIR检测方法的使用范围。 Objective:To observe the intervention effect of Danggui Tongfeng Formula on goutyarthritis(GA)rat synovium by Fourier transform infrared spectroscopy(FTIR),and to provide a new way for animal disease model determination and drug evaluation.Methods:A total of 60 SD rats were randomly divided into 6 groups:normal control group,model group,positive control group and low,medium and high groups of Danggui Tongfeng Formula,GA model was induced by intraperitoneal injection of potassium oxazate and sodium urate in knee joint cavity.One week later,Danggui Tongfeng Formula 13.08,26.16,52.32 g·kg^(-1)·d^(-1) was given by gavage,body weight,serum uric acid(UA),synovial fluid IL-6,synovial fluid IL-1β,pathological section,FTIR spectra and second derivative spectra were detected or observed.Results:Compared with the model group,the body weight on day 21 was significantly increased in high group of Danggui Tongfeng Formula(P<0.05),serum UA,synovial fluid IL-6 and IL-1βof the high group of Danggui Tongfeng Formula significantly decreased(P<0.05),and the pathological changes of synovium were improved.The FTIR spectra of the high group of Danggui Tongfeng Formula was close to that of the normal control group. Conclusion: For a certain extent, FIRT detection method can be used as one of the detection means to evaluate the efficacy of Danggui Tongfeng Formula in improving GA, which has important reference significance for the determination of GA animal models and drug evaluation, and expands the application range of FTIR detection method.
