瑶药穿心藤组培快繁体系的建立

Establishment of Tissue Culture and Rapid Propagation System of Yao Medicine Amydrium hainanense

摘要 目的:建立瑶药穿心藤高效组织培养再生体系,以利于保护和利用穿心藤种质资源。方法:以当年生穿心藤茎段为外植体,研究不同基础培养基对不定芽的诱导、不同浓度植物生长调节剂对不定芽诱导和分化以及不定芽增殖和生根的影响,筛选不同培养阶段的最适培养基。结果:最适穿心藤不定芽诱导培养基为改良MS+6-BA 2.00 mg/L+NAA 0.20 mg/L,诱导率达94.63%;最适不定芽增殖培养基为改良MS+6-BA 3.00 mg/L+NAA 0.40 mg/L,增殖系数可达10.30,芽绿且健壮;最适组培苗生根培养基为改良MS+NAA 0.50 mg/L,平均单株生根数为6条,生根率高达97.13%;组培苗炼苗6 d时,移栽苗长势健康,成活率最高,达100%。结论:建立了穿心藤的组织培养再生体系,为穿心藤种质资源保护和产业化种植奠定基础。 Objective:To establish an efficient tissue culture and regeneration system for the preservation and utilization of the germplasm resources of Yao medicine Amydrium hainanense.Methods:Using the stem segment of Amydrium hainanense in the current year as explants,the effects of different base medium on adventitious bud induction,plant growth regulator ratio of different concentration on adventitious bud induction and differentiation,bud proliferation and rooting were studied,and the optimal medium for different culture stages was screened.Results:The optimal medium for adventitive bud induction of Amydrium hainanense was MS,6-BA 2.00 mg/L and NAA 0.20 mg/L,the induction rate was 94.63%.The optimal medium for adventitious bud proliferation was MS,6-BA 3.00 mg/L and NAA 0.40 mg/L,the proliferation coefficient was up to 10.30,the buds were green and robust.The optimal medium for rooting of tissue culture seedlings was MS and NAA 0.50 mg/L,the average number of rooting per plant was 6,and the rooting rate was 97.13%.Trans-planting seedlings after 6 days of cultivation grew healthily and the survival rate was the highest,reaching 100%.Conclusion:The tissue culture and regeneration system of Amydrium hainanense is established,which can lay a foundation for the protection of germplasm re-sources and industrial planting of Amydrium hainanense.
