鸢尾苷元通过GSK-3β/Nrf2信号通路减轻Aβ_(1-42)诱导的SH-SY5Y细胞损伤 被引量:1

摘要 目的:探讨鸢尾苷元对β淀粉样蛋白(Aβ)_(1-42)诱导SH-SY5Y细胞损伤的保护作用及其潜在机制。方法:以Aβ_(1-42)诱导人神经母细胞瘤细胞系SH-SY5Y建立阿尔茨海默病(AD)神经元损伤模型,MTT检测细胞活力;流式细胞术检测细胞凋亡;相关试剂盒检测LDH、Caspase-3、氧化应激和GSK-3β水平;免疫荧光法检测4-HNE表达;PDB数据库和分子对接预测鸢尾苷元和GSK-3β的结合;Western Blot分析凋亡和GSK-3β/Nrf2通路相关蛋白表达。结果:鸢尾苷元可呈浓度依赖性缓解Aβ_(1-42)诱导的SH-SY5Y细胞活力损伤、凋亡及氧化应激,并可通过靶向GSK-3β从而调控GSK-3β/Nrf2信号。结论:鸢尾苷元可通过调控GSK-3β/Nrf2通路进而抑制Aβ_(1-42)诱导的SH-SY5Y细胞损伤。
