BDNF上游调控机制及表达水平与精神分裂症认知功能损伤的相关性 被引量:1

Correlation of upstream regulatory mechanisms and expression levels of BDNF with cognitive impair-ment in schizophrenia

摘要 目的探讨精神分裂症患者外周血脑源性神经营养因子(Brain-Derived Neurotrophic Factor,BDNF)表达水平与其认知功能损伤的相关性。方法选择符合诊断标准的精神分裂症患者54例,健康对照55例,采用阳性与阴性症状量表(Positive and Negative Syndrome Scale,PANSS)评估精神分裂症患者的临床症状,采用精神分裂症认知功能成套测验(MATRICS Consensus Cognitive Battery,MCCB)评估受试者的认知功能,采用逆转录实时荧光定量聚合酶链式反应检测受试者外周血microRNA124-3p及脑源性神经营养因子的信使RNA(Messenger RNA for Brain-Derived Neurotrophic Factor,BDNFmRNA)相对表达量,采用酶联免疫分析检测受试者外周血BDNF浓度。结果1.与对照组相比,患者组microRNA124-3p表达量升高(P<0.001),BDNFmRNA和BDNF表达量降低(P<0.001),在MCCB中的信息处理速度、词语学习、推理和问题解决、视觉学习四个方面明显下降(P<0.001)。2.在患者组中,BDNFmRNA相对表达量是PANSS总分的正向影响因素(P<0.05),microRNA124-3p相对表达量是阳性症状得分的负向影响因素(P<0.05)。3.在患者组中,microRNA124-3p相对表达量是推理和问题解决(Reasoning and Problem Solving,RPS)得分的负向影响因素(P<0.05),BDNF蛋白因子浓度是RPS得分的正向影响因素(P<0.05);在对照组中,BDNF蛋白因子浓度是RPS得分的正向影响因素(P<0.05)。结论精神分裂症患者外周血BDNF表达量与其认知损伤严重程度负相关。 Objective To investigate the correlation between peripheral blood Brain-Derived Neurotrophic Factor(BDNF)expression levels and their cognitive impairment in patients with schizophrenia.Methods 54 schizophrenia patients meeting the criteria and 55 healthy controls were selected.The clinical symptoms of schizophrenia patients were assessed using the Positive and Negative Syndrome Scale(PANSS),and the cognitive function of the subjects was assessed using MATRICS Consensus Cognitive Battery(MCCB).The relative expression of peripheral blood microRNA124-3p and Messenger RNA for Brain-Derived Neurotrophic Factor(BDNFmRNA)were measured by Reverse Transcription-Polymerase Chain Reaction,and the concentration of BDNF in peripheral blood was measured by enzyme-linked immunoassay.Results 1.Compared with the control group,the patient group showed elevated microRNA124-3p expression(P<0.001),decreased BDNFmRNA and BDNF expression(P<0.001),and significantly decreased in four areas of information processing speed,word learning,reasoning and problem solving,and visual learning in MCCB(P<0.001).2.In the patient group,the relative expression of BDNFmRNA was a positive influence on the total PANSS score(P<0.05)and the relative expression of microRNA124-3p was a negative influence on the positive symptom score(P<0.05).3.In the patient group,the relative expression of microRNA124-3p was a negative influence on the Reasoning and Problem Solving(RPS)score(P<0.05)and BDNF protein factor concentration was a positive influence on the RPS score(P<0.05).In the control group,BDNF protein factor concentration was a positive influence on the RPS score(P<0.05).Conclusion Peripheral blood BDNF expression in schizophrenia patients was negatively correlated with the severity of their cognitive impairment.
