摘要
目的利用蛋白质组学技术筛选精神分裂症患者血清中表达差异的蛋白质,以探讨精神分裂症致病机制。方法收集13例精神分裂症患者和6例健康志愿者(对照组)血清样本,采用非标记蛋白质组学技术联合液相色谱-质谱分析技术鉴定两组的差异蛋白,筛选出表达差异显著的蛋白质,并进行基因本体和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析。结果非标记蛋白质组学结果显示精神分裂症患者血清中共筛选出126个显著性差异的蛋白,其中63个上调蛋白,63个下调蛋白。这些差异蛋白主要富集在血液微粒、细胞质囊腔、囊腔、分泌颗粒管腔等细胞组分,分子功能主要表现在肽酶调节活性,主要参与蛋白激活级联、体液免疫应答、血小板脱粒、补体激活、蛋白修饰、急性炎症反应等生物学过程。精神分裂症主要富集的KEGG通路包括补体和凝血级联反应、冠状病毒和全身性红斑狼疮。结论精神分裂症患者血清中差异蛋白主要富集在补体和凝血级联反应通路,揭示该通路在精神分裂症发病过程中发挥重要作用。
Objective The aim of the present study was to screen the differentially expressed proteins in the serum of patients with Schizophrenia by proteomics technology,and to explore the pathogenic mechanism of schizophrenia.Methods Serum samples were collected from 13 Schizophrenia patients and 6 healthy volunteers(Control group).The differentially expressed proteins between Schizophrenia group and Control group were identified by labelfree proteomics via liquid chromatography-mass spectrometry.The differentially expressed proteins were analyzed by gene ontology and KEGG(Kyoto Encyclopedia of Genes and Genomes)enrichment pathway analysis.Results Label-free proteomics results showed that a total of 126 differentially expressed proteins were screened between Schizophrenia group and Control group,including 63 up-regulated proteins and 63 down-regulated proteins.These differentially expressed proteins were mainly enriched in cellular components such as blood microparticle,cytoplasmic vesicle lumen,vesicle lumen,secretory granule lumen;and were mainly involved in biological processes such as protein activation cascade,humoral immune response,platelet degranulation,complement activation,protein processing and acute inflammatory response.The molecular function is peptidase regulatory activity.The KEGG pathways enrichment analysis showed that the differentially proteins were mainly enriched complement and coagulation cascades,coronavirus disease,systemic lupus erythematosus.Conclusion The differentially proteins in the serum of schizophrenia patients were mainly enriched in the complement and coagulation cascades,which revealed that the pathway play an important role in the pathogenesis of schizophrenia.
