摘要
目的分析首发精神分裂症与健康人群血浆外泌体的差异表达蛋白,筛选出临床早期诊断等密切相关的生物学标志物。方法收集15例首发精神分裂症患者和45例健康体检者的血浆,提取两组标本的外泌体蛋白质。采用定量蛋白质组学串联质谱标签(Tandem mas tags,TMT)标记技术进行蛋白质定量分析,并利用MaxQuant软件、UniProt人类蛋白组SP数据库和在线工具String对差异蛋白进行生物信息学分析。结果本实验以健康人群为参照,首发患者鉴定并筛选出98个差异蛋白,92个表达上调,6个表达下调。初步筛选出可能与精神分裂症发生发展相关的热休克蛋白90α(Heat shock protein HSP 90-alpha,HSP90AA1)、蛋白酶体亚型β1和β2(Proteasome subunit beta 1,2,PSMB1和PSMB2)共3个候选蛋白标志物。结论首发精神分裂症存在差异表达的蛋白,有可能成为精神分裂症发生发展相关的蛋白标志物。
Objective The differentially expressed plasma exosome proteins of first episode schizophrenic patients and healthy people was analyzed to screen out biomarkers closely related to early clinical diagnosis.Method Plasma samples of 15 patients with first episode schizophrenia and 45 healthy subjects were collected and secreted proteins of the two groups were extracted.TMT connecting with Mass spectrometry technique was used for protein quantitative analysis,and Maxquant software,Uniport databae and String website were used for bioinformatics analysis of differential proteins.Results Ninety-eight differentially expressed plasma exosome proteins were identified inthe first episode schizophrenic patients compared with the healthy people,92 proteins were significantly up-regulated,and 6 proteins were significantly down-regulated(P<0.01).HSP90AA1,Proteasome subunit beta 1 and Proteasome subunit beta 2 were preliminarily screened as candidate protein markers related to schizophrenia.Conclusion Differentially expressed proteins in first-episode schizophrenia may become the protein markers related to the occurrence and development of schizophrenia.
