非小细胞肺癌患者血管性血友病因子和微小RNA-497表达水平与肿瘤血管生成关系

Relationship between the expression levels of von Willebrand factor and miRNA-497 and tumor angiogenesis in patients with non-small cell lung cancer

摘要 目的:探究血管性血友病因子(vWf)和微小RNA-497(miRNA-497)在非小细胞肺癌(NSCLC)患者中的表达情况及与肿瘤血管生成的关系。方法:收集102例接受手术根治治疗的NSCLC患者,免疫组化法测定患者癌变组织和对应癌旁组织(正常组织)中vWf表达情况和微血管密度(MVD),实时荧光定量聚合酶链反应(PCR)检测微小RNA-497(miRNA-497)表达情况。比较不同组织中vWf、miRNA-497表达水平,探究其表达水平与NSCLC患者MVD的关系。结果:癌变组织中vWf表达水平和MVD计数明显高于癌旁组织,miRNA-497表达水平明显低于癌旁组织(均P<0.05)。vWf在TNMⅢ-Ⅳ期、肿瘤细胞中低分化及出现淋巴转移的NSCLC癌变组织中表达水平明显提高,miRNA-497在该类患者癌变组织中表达水平明显下降(均P<0.05)。相关性分析显示,NSCLC癌变组织中vWf表达水平与MVD计数呈正相关(r=0.554),miRNA-497表达水平与MVD计数呈负相关(r=-0.707,P<0.01)。结论:vWf在NSCLC病灶组织中上调表达,miRNA-497下调表达,且其表达均与肿瘤细胞分化、侵袭、转移和血管新生有关。 Objective:To investigate the expression of von Willebrand factor(vWf)and miRNA-497 in patients with non-small cell lung cancer(NSCLC)and their relationship with tumor angiogenesis.Methods:102 patients with NSCLC who underwent radical surgical treatment were collected.The expression of vWf and microvessel density(MVD)in the cancerous tissues and corresponding adjacent tissues(normal tissues)were determined by immunohistochemistry,and real-time fluorescence quantitative polymerase chain reaction(PCR)was used to determine miRNA-497 expression in lung tissue.The expression levels of vWf and miRNA-497 in different tissues were compared,and which relationship between with MVD in NSCLC patients was analyzed.Results:Compared with the expression level of the adjacent tissues of NSCLC patients,the expression level of vWf in the cancerous tissues was increased,the number of MVD was increased,and the level of miRNA-497 was decreased(all P<0.05).The expression level of vWf was significantly increased in TNM Ⅲ to Ⅳ stage,poorly differentiated NSCLC tissues and lymphatic metastases,and the expression level of miRNA-497 was significantly decreased in these patients(all P<0.05).Correlation analysis showed that there was a positive correlation between the expression of vWf in NSCLC cancerous tissues and the number of MVD,while the expression level of miRNA-497 was negatively correlated with MVD count(r=0.554,-0.707,all P<0.01).Conclusion:The expression of vWf is up-regulated,and miRNA-497 down-regulated in NSCLC focal tissues,and its expression is correlated with tumor cell differentiation,invasion,metastasis and angiogenesis.
