摘要
为了解长春地区犬猫源金黄色葡萄球菌(Staphylococcu saureus,S.aureus)和伪中间型葡萄球菌(Staphylococcus pseudintermedius,SP)耐药基因和毒力基因携带情况,利用高通量测序技术对17株S.aureus与38株SP所携带的耐药基因与毒力基因进行分析,根据Kirby-Bauer纸片扩散法的结果分析耐药基因型与表型的关系,同时进行了MLST分型。结果表明S.aureus和SP对常规抗菌药物表现中介或耐药,但对呋喃妥因敏感率为100%;β-内酰胺类耐药基因bla_(Z)检出率较高,分别为100%和57.9%;各有1株同时携带bla_(Z)和bla_(NDM);S.aureus中检出2株携带磷霉素类耐药基因fosA;SP中检出21株携带多药耐药基因cfr;各检出3株携带多药外排耐药基因mdfA;共检出4株MRSA和11株MRSP;优势ST型分别为ST25和ST1430;在大多数分离菌中携带有γ-溶血素hlgB、黏附素clf、fnb、杀白细胞素LukF-PV、LukS-PV等毒力基因。分离株耐药性严重。研究结果可为兽医临床中治疗凝固酶阳性葡萄球菌感染用药提供数据支持。
In order to understand the carrying situation of drug resistance genes and virulence genes of Staphylococcus aureus(S.aureus)and Staphylococcus pseudintermedius(SP)from dogs and cats in Changchun,high throughput sequencing technology was used to analyze the drug resistance genes and virulence genes carried by 17 strains of S.aureus and 38 strains of SP.According to results of Kirby-Bauer disk diffusion method,the relationship between drug resistance genotype and phenotype was analyzed,and MLST typing was carried out.The results showed that S.aureus and SP were intermediate or resistant to conventional antibiotics,but the sensitivity to nitrofurantoin was 100%.The detection rates ofβ-lactam resistance gene bla_(Z)were 100%and 57.9%,respectively.One of S.aureus and one of SP both carried bla_(Z)and bla_(NDM).Two strains of S.aureus carrying fosfomycin resistance gene fosA were detected.Twenty-one strains carrying multidrug resistance gene cfr were detected in SP.Three strains carrying multidrug efflux resistance gene mdfA were detected.A total of 4 strains MRSA and 11 strains of MRSP were detected.The dominant ST types were ST25 and ST1430.Most of the isolated bacteria carried virulence genes such asγ-hemolysin hlgB,adhesin clf,fnb,leucocidin LukF-PF,and LukS-PV.The isolates were highly drug resistant.The results can provide data support for the treatment of coagulase-positive staphylococcal infection in veterinary clinic.
