气相色谱法测定菜籽油中角鲨烯的含量

Determination of Squalene Content in Rapeseed Oil by Gas Chromatography

目的:建立一种菜籽油中角鲨烯的气相色谱方法。方法:样品经氢氧化钾-甲醇溶液皂化后,用正己烷提取,经HP-5色谱柱分离,气相色谱仪分析测定,外标法定量。结果:角鲨烯在10~100 mg·L^(-1)呈现良好的线性关系,相关系数为0.999 8,回收率为91.0%~98.7%,相对标准偏差为3.2%~6.5%,检出限为1 mg·kg^(-1),定量限为3.5 mg·kg^(-1)。结论:该方法的线性良好,结果准确,精密度高,适用于菜籽油中角鲨烯含量的测定。

Objective:To establish a gas chromatography method for squalene in rapeseed oil.Method:The samples were saponified by potassium hydroxide and methanol solution,extracted with n-hexane,separated by HP-5 column,analyzed by gas chromatograph,and quantified by external standard method.Result:At 10~100 mg·L^(-1),squalene showed a good linear relationship,the correlation coefficient was 0.9998,the recovery was 91.0%~98.7%,the relative standard deviation was 3.2%~6.5%,the limit of detection was 1 mg·kg^(-1),and the limit of quantification was 3.5 mg·kg^(-1).Conclusion:The method has good linearity,accurate results and high precision,and is suitable for the determination of squalene content in rapeseed oil.
