小鼠自身给药模型颈静脉插管手术改进

The improvement of the mouse jugular vein cannulation surgery used in self⁃administration model

摘要 目的自身给药模型是药物成瘾研究中重要的动物模型之一,实验动物静脉插管手术是自身给药模型构建是否成功的关键步骤。本研究旨在改进小鼠颈静脉插管手术和构建小鼠甲基苯丙胺自身给药模型。方法改进小鼠颈静脉插管手术方法,将改进后的一体化的生物垫片和导管插入40只C57BL/6J小鼠的侧颈部静脉内;待小鼠颈静脉手术恢复后,进行甲基苯丙胺自身给药训练。结果小鼠术后恢复良好,导管保持畅通,手术存活率达到92.5%。共计35只小鼠(87.5%)进入自身给药训练,33只小鼠(94.3%)自身给药模型构建成功。结论改进小鼠颈静脉插管手术,可较好保证小鼠颈静脉插管的操作性和畅通性,促进小鼠甲基苯丙胺自身给药模型成功构建,为开展小鼠自身给药实验提供良好的技术基础。 Objective Self⁃administration is one of the widely⁃used animal models in the study of drug addiction.Intravenous cannula surgery is a key step in the success of the acquisition and maintenance of self⁃administration behavior in the animals.Methods By improving the surgical method of jugular vein cannulation,the integrated biological spacer and catheter was inserted into the lateral neck vein of 40 C57BL/6J mice.Methamphetamine self⁃administration training was carried out after the animals recovered from surgery.Results Mice recovered well after surgery and the catheter remained patent.The survival rate reached 92.5%.A total of 35 mice(87.5%)were trained to self⁃administer methamphetamine following recovery.A total of 33 mice(94.3%)exhibited stable methamphetamine self⁃administration behavior.Conclusion Improving the jugular cannularization of mice can better ensure the quality of the surgery and the patency of the jugular cannula in mice.It greatly helps successful training and maintenance of methamphetamine self⁃administration behavior in mice.
