饲料用连翘叶不同提取物体外抗炎抑菌活性的比较研究

摘要 研究旨在比较饲料用连翘叶不同提取物的体外抗炎活性和抑菌活性。运用脂多糖诱导的小鼠单核巨噬细胞RAW264.7研究连翘叶水提取物、连翘叶40%乙醇提取物和连翘叶80%乙醇提取物的体外抗炎活性,并通过抑菌试验对3种连翘叶提取物进行了体外抑菌活性的比较研究。采用Griess法检测一氧化氮(NO)生成水平,酶联免疫分析法(ELISA)检测白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的生成水平。并使用纸片扩散法和微量肉汤倍比稀释法测定抑菌活性、最低抑菌浓度和最低杀菌浓度,结果表明:3种连翘叶提取物均能抑制NO、IL-1β、IL-6和TNF-α的生成,抗炎活性80%乙醇提取物>40%乙醇提取物>水提取物。抑菌活性40%乙醇提取物>水提取物>80%乙醇提取物,3种连翘叶提取物对金黄色葡萄球菌的抑菌作用最强。饲料用连翘叶乙醇提取物具有较好的体外抗炎活性和抑菌作用,可作为新型饲料添加剂开发应用。 The study was to compare the anti-inflammatory and antibacterial activities of feed different extracts of Forsythia suspensa leaves in vitro.The mouse mononuclear macrophages RAW264.7 induced by lipopolysaccharide were used to compare the anti-inflammatory activity of aqueous extract,40%ethanol extract and 80%ethanol extract of Forsythia suspensa leaves in vitro.The antibacte-rial activities of three extracts of Forsythia sus-pensa leaves were compared by antibacterial ex-periment in vitro.The level of nitric oxide(NO)production was measured by Griess method,and the levels of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-a(TNF-a)were measured by enzyme-linked immunosorbent assay(ELISA).The antibacterial activity,minimum inhibitory concentration and minimum bactericidal concentration were measured by disk diffusion method and microdilution of broth double dilution method.The results showed that the three extracts of Forsythia suspensa leaves could inhibit the production of NO,IL-1β,IL-6 and TNF-a.The anti-inflammatory activity were 80%ethanol extract>40%ethanol extract>aqueous extract.The antibacterial activity were 40%ethanol extract>aqueous extract>80%ethanol extract.Three extracts of Forsythia suspensa leaves showed the strongest antibacterial effect against Staphylococcus aureus.The feed ethanol extract of Forsythia suspensa leaves has better anti-inflammatory and antibacterial activities in vitro,which provides experimental basis for the effective development of Forsythia suspensa leaves as new feed additives.
