摘要
开展花生茎叶中黄酮类化合物的高效分离与活性分析研究。采用高速逆流色谱技术进行分离纯化,并结合分子对接技术评价其助睡眠活性。筛选最佳溶剂体系为正己烷/乙酸乙酯/甲醇/水(5∶5∶2∶8,5∶5∶3∶7,5∶5∶5∶5,v/v),通过梯度洗脱分离得到两种化合物,经质谱和核磁结构鉴定为大豆苷元和芒柄花黄素。采用分子对接分析大豆苷元和芒柄花黄素对失眠相关的两种蛋白受体(GABRA1和FOS)均有较好的亲和力,相互作用力主要为氢键,两种化合物具有助睡眠的潜在活性。建立了花生茎叶中大豆苷元和芒柄花黄素的高速逆流色谱分离方法,采用分子对接证实两种化合物具有助睡眠的潜在活性,为花生茎叶中化学成分的分离及进一步开发助睡眠活性化合物提供了参考。
Conduct efficient separation and activity analysis of flavonoids in peanut stems and leaves.The high-speed countercurrent chromatography was used for flavonoids separation and purification,and molecular docking technology was used to evaluate their effect on sleep.The optimum separation solvent system was n-hexane/ethyl acetate/methanol/water(5∶5∶2∶8,5∶5∶3∶7,5∶5∶5∶5,v/v).Two compounds were separated by gradient elution.They were identified as daidzein and formononetin by MS and NMR.Both daidzein and formononetin had good affinity for two protein receptors related to insomnia(GABRA1 and FOS),with hydrogen bonding as the main interaction force.Two compounds had potential sleep aid activities.The high-speed countercurrent chromatography was established for separation of daidzein and formononetin in peanut stems and leaves.These two compounds had potential sleep aid activities evaluated by molecular docking technology.This study provides a reference for the separation of chemical components from peanut stems and leaves and the further study of sleep aid active compounds.
