ICR鼠肝和肾毒性损伤生物标志物的筛选

摘要 肝脏和肾脏是生物体受到外界毒性刺激最易损伤的器官,ICR小鼠又是动物乃至人类病理学评估的重要模式生物。为筛选可用于ICR鼠肝和肾毒性损伤的生物标志物,采用来自受污染的北方两个不同矿区水样对ICR小鼠进行灌胃,建立小鼠肝、肾毒性损伤模型,将其样本进行高通量测序,利用转录组数据进行收集、分组并对比分析,同时采用体外肾细胞NRK的毒性试验进行验证,最终筛选可用于毒性损伤的分子标记物。结果表明,基因Chordc1与Cry1可作为ICR鼠肝和肾毒性损伤生物标志物。试验结果可为有毒有害物对于生物体肝、肾损伤的诊断和健康评价提供一种新思路。 The liver and kidneys are highly susceptible to external toxic stimuli.For the evaluation of animal and human pathology,ICR mice serve as important model organisms.To screen for biomarkers of liver and kidney toxicity injury in ICR mice,samples of water from two mining areas in northern China were used to create a model of toxicity injury through gavage.The resulting samples were then subject to high-throughput sequencing,followed by data collection,grouping,and analysis of the transcriptome.Molecular markers for the toxicity injury were screened by validating them through renal NRK toxicity in vitro.Finally,molecular markers for toxicity injury were screened.The study reveals that Chordc1 and Cry1 are viable biomarkers for liver and kidney toxicity injury in ICR mice.The findings offer novel perspectives on the identification and clinical evaluation of liver and kidney injury in living organisms caused by exposure to poisonous or hazardous substances.
