APOE基因多态性对脑网络衰老变化模式的影响

摘要 目的通过纵向研究,消除个体之间的差异,更清楚地表征APOEε4基因在大脑衰老中的作用。方法该研究纳入26例APOE4+,26例APOE4-志愿者,采用数据驱动的方法,采用静态功能连接对APOEε4携带者和非携带者进行随访。结果静态功能连接发现:网络内功能连接差异主要位于DMN网络(左侧后扣带回和双侧楔前叶)。结论尽管神经心理学量表未表现出差异,但是静态功能连接已发生改变。这有可能解释APOEε4等位基因加速生物衰老的假说,为探索正常衰老和AD提供新的研究思路。 Objective To eliminate the differences among individuals and better characterize the role of APOEε4 gene in brain aging through the longitudinal study.Methods This study included 26 APOE4+and 26 APOE4 volunteers,and adopted a data-driven approach to follow up APOEε4 carriers and non-carriers in combination with static functional connections.Results Static functional connectivity was found:the differences in within-network functional connectivity were mainly located in the DMN network(left posterior cingulate gyrus and bilateral precuneus).Conclusion Although the neuropsychological scale showed no differences,the static functional connections were altered.This may explain the hypothesis that APOEε4 allele accelerates biological aging,providing a new research approach for exploring normal aging and AD.
