复方甘乐注射液对小鼠血糖的影响研究

摘要 目的:研究复方甘乐注射液对小鼠血糖的影响。方法:选健康雄性KM小鼠70只,随机分为7组,分别为复方甘乐注射液高剂量组(416 mg/kg)、中剂量组(208 mg/kg)、低剂量组(104 mg/kg)、左卡尼汀注射液组(200 mg/kg)、甘乐注射液剂量组(8 mg/kg)、胰岛素注射液组(0.8 U/kg)及空白对照组(氯化钠注射液)。皮下给予相应受试物7天,比较给药后的血糖值。建立高血糖小鼠模型,将90只糖尿病模型小鼠随机分为9组,分别为复方甘乐注射液高剂量组(416 mg/kg)、中剂量组(208 mg/kg)、低剂量组(104 mg/kg),左卡尼汀注射液高剂量组(400 mg/kg)、低剂量组(200 mg/kg),甘乐注射液高剂量组(16 mg/kg)、低剂量组(8 mg/kg),胰岛素注射液组(0.8 U/kg)及模型对照组。同时取10只正常小鼠作为空白对照组(氯化钠注射液)。皮下给予相应受试物7天,比较给药后的血糖值。建立高血糖小鼠模型,将80只糖尿病模型小鼠随机分为8组,分别为复方甘乐注射液组(208 mg/kg)、复方甘乐注射液(208 mg/kg)+胰岛素注射液组(0.4 U/kg)、左卡尼汀注射液组(200 mg/kg)、左卡尼汀注射液(200 mg/kg)+胰岛素注射液组(0.4 U/kg)、甘乐注射液组(8 mg/kg)、甘乐注射液(8 mg/kg)+胰岛素注射液组(0.4 U/kg)、胰岛素注射液组(0.4 U/kg)及模型对照组,同时取10只正常小鼠作为空白对照组(氯化钠注射液)。皮下给予相应受试物7天,比较给药后的血糖值。结果:复方甘乐注射液各剂量组对正常小鼠血糖无显著影响。高血糖模型中,复方甘乐注射液中剂量组(208 mg/kg)、左卡尼汀注射液组(200 mg/kg)、甘乐注射液组(8 mg/kg)及胰岛素注射液组(0.8 U/kg)与模型组比较,差别有统计学意义(P<0.05)。复方甘乐注射液(208 mg/kg)+胰岛素注射液组(0.4 U/kg)、左卡尼汀注射液(200 mg/kg)+胰岛素注射液组(0.4 U/kg)、甘乐注射液(8 mg/kg)+胰岛素注射液组(0.4 U/kg)与模型组比较,差别有统计学意义(P<0.01);并且复方甘乐注射液(208 mg/kg)+胰岛素注射液组(0.4 U/kg)与胰岛素注射液组(0.4 U/kg)比较,差别有统计学意义(P<0.01)。结论:在本实验条件下,复方甘乐注射液对正常小鼠血糖未见有调节作用,对四氧嘧啶糖尿病模型小鼠血糖具有一定的调节作用。 Objective:To study the effect of compound Ganle injection on blood glucose in mice.Method:Seventy healthy male KM mice were selected and randomly divided into seven groups:high-dose group(416 mg/kg),medium dose group(208 mg/kg),low-dose group(104 mg/kg),levocarnitine injection group(200 mg/kg),Ganle injection dose group(8mg/kg),insulin injection group(0.8 U/kg),and blank control group(sodium chloride injection).Subcutaneously administer the corresponding test substance for 7 days and compare the blood glucose levels after administration.To establish a hyperglycemic mouse model,90 diabetes model mice were randomly divided into 9 groups:high dose group(416 mg/kg),medium dose group(208 mg/kg),low dose group(104 mg/kg),high dose group(400 mg/kg),low dose group(200 mg/kg),high dose group(16 mg/kg),low dose group(8 mg/kg)Insulin injection group(0.8 U/kg)and model control group.Simultaneously,10 normal mice were taken as the blank control group(sodium chloride injection).Subcutaneously administer the corresponding test substance for 7 days and compare the blood glucose levels after administration.To establish a hyperglycemic mouse model,80 diabetes model mice were randomly divided into 8 groups:Compound Ganle injection Study on the Effect of Compound Ganle Injection on Blood Glucose in Mice group(208 mg/kg),Compound Ganle injection group(208 mg/kg)+insulin injection group(0.4 U/kg),L-carnitine injection group(200 mg/kg),L-carnitine injection group(200 mg/kg)+insulin injection group(0.4 U/kg),Ganle injection group(8 mg/kg)Ganle injection(8 mg/kg)+insulin injection group(0.4 U/kg),insulin injection group(0.4 U/kg)and model control group,while taking 10 normal mice as blank control group(sodium chloride injection).Subcutaneously administer the corresponding test substance for 7 days and compare the blood glucose levels after administration.Result:Each dose group of Compound Ganle Injection had no significant effect on blood glucose levels in normal mice.In the hyperglycemic model,there was a significant difference(P<0.05)between the medium dose group of compound Ganle injection(208 mg/kg),the group of levocarnitine injection(200 mg/kg),the group of Ganle injection(8 mg/kg),and the group of insulin injection(0.8 U/kg)compared to the model group.Compound Ganle Injection(208 mg/kg)+Insulin Injection Group(0.4 U/kg),Levocarnitine Injection(200 mg/kg)+Insulin Injection Group(0.4 U/kg),Ganle Injection(8 mg/kg)+Pancreatic Islet.The difference between the Su injection group(0.4 U/kg)and the model group was statistically significant(P<0.01);Moreover,there was a statistically significant difference(P<0.01)between the compound Ganle injection(208 mg/kg)+insulin injection group(0.4 U/kg)and insulin injection group(0.4 U/kg).Conclusion:Under the experimental conditions,Compound Ganle Injection has no regulating effect on blood glucose in normal mice,but has a certain regulating effect on blood glucose in alloxan diabetes model mice.
