豆蔻酰化在肿瘤治疗中的研究进展

摘要 N-豆蔻酰化是一种蛋白质修饰方式,也是常见的蛋白质脂化类型之一。N-豆蔻酰化通过改变蛋白构象、稳定性及细胞定位,影响细胞信号转导、代谢途径重编程以及参与各种生理及病理过程。研究表明N-豆蔻酰化转移酶(NMT)在各类恶性肿瘤中异常表达参与肿瘤发生、发展。因此,靶向NMT抑制剂具有良好抗癌效果,为癌症复发及转移治疗提供了潜在靶点。文章主要介绍了蛋白质N-豆蔻酰化系统并综述其在肿瘤发生、发展中的作用及机制,总结近5年靶向蛋白质N-豆蔻酰化治疗癌症的最新研究进展。 N-myristoylation is a type of protein modification and one of the most common types of protein lipidation.N-myristoylation affects cell signal transduction and metabolic pathway reprogramming by changing protein conformation,stability and localization.It is involved in various physiological and pathological processes.Studies have shown that the protein N-myristoyltransferase(NMT)is abnormally expressed in many types of malignancies and involves in development and progression of tumors.Inhibitors targeting NMT have good anticancer effects and provide potential targets for cancer recurrence and metastasis therapy.This article mainly introduces the protein N-myristoylation system and reviews its role and mechanism in the development and progression of tumors,summarizes the latest research progress of targeting protein N-myristoylation for the treatment of cancer in the last 5 years.
