罗汉果根的生物活性及化学成分

Chemical Constituents from the Root of Siraitia grosvenorii and Their Biological Activities

摘要 以受根结线虫危害的罗汉果根为研究对象,研究不同极性部位的生物活性及化学成分。罗汉果根粗提物过HPD-100大孔树脂层析柱,依次用不同浓度甲醇梯度洗脱,收集洗脱液,得到Fr1,Fr2,Fr3,Fr4部位,利用传统分离方法和反相制备色谱技术进行分离纯化,采用ESI-MS,NMR对化合物进行结构鉴定;以抑制α-葡萄糖苷酶和HepG2肿瘤细胞能力为指标,研究各极性部位和部分化合物的生物活性。结果表明:Fr4部位具有最强的抑制α-葡萄糖苷酶和HepG2肿瘤细胞增殖活性。从Fr4中分离得到4种葫芦素,分别为双氢葫芦素E(1),葫芦素E(2),二氢异葫芦素B-25-乙酸酯(3),葫芦素B(4),化合物均为首次从罗汉果属中分离得到,且化合物4有较好的抗肿瘤活性。 To study the biological activities and chemical constituents of Siraitia grosvenori roots,the crude extracts of S.grosvenori roots were loaded to HPD-100 column and eluted by gradient methanol-water solutions to obtain Frl,Fr2,Fr3 and Fr4 fractions.Traditional separation methods and reversed-phase preparative chromatography techniques were used for separation and purification,and ESI-MS and NMR were used for identifying their structure.α-glycosidase and HepG2 tumor cell inhibition activity of different polar parts and partical compounds were determined.The results indicate that Fr4 has the highestα-glycosidase and HepG2 tumor cell inhibition activity.Four compounds were isolated from Fr4,and their structures were identified as dihydrocucurbitacin E,cucurbitacin E,dihydroisocucurbitacin B-25-acetate and cucurbitacin B,and they are isolated from S.grosvenorii for the first time.
