响应面法优化叉分蓼多糖的提取及其结构分析 被引量:1

Optimization of extraction of Polygonum divaricatum L.polysaccharides by response surface methodology and its structural analysis

摘要 采用响应面法优化叉分蓼多糖的水提工艺,并采用紫外光谱(UV)、红外光谱(FT-IR)、粒径与Zeta电位仪、刚果红试验及扫描电子显微镜(SEM)对其结构进行分析。结果表明:在料液比1∶40(g/mL)、浸提温度95℃、浸提时间4 h的最佳工艺条件下,多糖得率达7.31%±0.34%。UV分析结果显示,脱蛋白质后的叉分蓼多糖几乎不含蛋白质及核酸等物质。FT-IR分析结果表明,叉分蓼多糖存在多糖特征吸收信号,平均粒径为227.96 nm,平均Zeta电位为-21.92 mV。刚果红试验推测,叉分蓼多糖具有螺旋结构,其微观结构表现为无规则海绵状,表面含许多空腔。 The response surface methodology was used to optimize the water extraction process of Polygonum divaricatum L.polysaccharides,and their structures were analyzed using ultraviolet spectroscopy(UV),infrared spectroscopy(FT-IR),particle size and Zeta potential analyzer,Congo red assay,and scanning electron microscopy(SEM).The results showed that under the optimal process conditions of a solid-liquid ratio of 1:40(g/mL),extraction temperature of 95℃,and extraction time of 4 h,the yield of polysaccharides reached 7.31%±0.34%.The results of UV analysis indicated that it contained almost no substances such as proteins or nucleic acids after deproteinization.The results of FT-IR analysis indicated that there was a polysaccharide characteristic absorption signalin Polygonum divaricatum L.polysaccharide.The average particle size was 227.96 nm,and the average Zeta potential was -21.92 mV.The Congo red test speculated that the Polygonum divaricatum L.polysaccharide had a helical structure.The microstructure exhibited an irregular sponge like structure with many cavities on the surface.
