GP1BA、PTGS1、LTC4S、ITGB3基因多态性检测指导阿司匹林使用

GP1BA,PTGS1,LTC4S and TGB3 gene polymorphism testing for guiding aspirin use

目的对陕西宝鸡地区阿司匹林药物相关基因的基因型分布进行回顾性分析,明确与阿司匹林药物相关基因的突变率。方法选取2022年1月至2023年5月在宝鸡市中医医院心内科接受阿司匹林治疗并进行相关基因检测的住院患者475例,采用飞行时间质谱仪对阿司匹林药物相关的4个基因[血小板膜糖蛋白Ⅰbα多肽(GP1BA)基因、前列腺素内过氧化物合酶1(PTGS1)基因、白三烯C4合酶(LTC4S)基因、糖蛋白Ⅲa(ITGB3)基因]进行检测,然后进行基因多态性分析。结果各基因分布频率符合Hardy-Weinberg平衡定律,GP1BA c.482C>T基因型分布频率CC>CT>TT,ITGB3 c.176T>C基因型分布频率TT>TC>CC,PTGS1 c.-842A>G基因型分布频率AA>AG>GG,LTC4S c.-444A>C基因分布频率AA>AC>CC;不同性别、年龄的各基因型分布比较,差异无统计学意义(P>0.05)。结论对阿司匹林药物治疗患者进行阿司匹林药物相关基因检测,可以为临床医生对患者制订个体化治疗方案提供辅助性的指导,具有重要的临床价值。

Objective To retrospectively analyze the distribution of genotypes of aspirin drug-related genes in Baoji area of Shaanxi,and to definite the mutation rate of aspirin drug-related genes.Methods A total of 475 inpatients receiving aspirin treatment in the cardiologic medicine department of Baoji Municipal Hospital of Traditional Chinese Medicine from January 2022 to May 2023 were selected.The four genes related to aspirin drugs(GP1BA gene,PTGS1 gene,LTC4S gene,ITGB3 gene)were detected by the time-of-flight mass spectrometer,and then the gene polymorphism of the detection results was analyzed.Results The distribution frequency of each gene was in line with Hardy-Weinberg equilibrium rule,GP1BA c.482C>T genotype distribution frequency CC>CT>TT,ITGB3 c.176T>C genotype distribution TT>TC>CC,PTGS1 c.-842A>G genotype distribution frequency AA>AG>GG,LTC4S c.-444A>C genotype distribution AA>AC>CC.There was no statistically significant difference in genotype distribution between the different genders and ages.Conclusion Aspirin drug-related gene testing for the patients with aspirin drug treatment could provide the auxiliary guidance for clinicians to formulate the individualized treatment regimen for the patients,which has an important clinical value.
