SDF-1/CXCR4信号轴在MSCs修复损伤组织中作用的研究进展

摘要 骨髓间充质干细胞(mesenchymal stem cells,MSCs)具有自我更新和多向分化潜能,在损伤组织修复中起着重要作用。基质细胞衍生因子-1(stromal cell-derived factor-1,SDF-1)/CXC趋化因子受体4(CXC chemokine receptor 4,CXCR4)信号轴是由SDF-1与其受体CXCR4相互作用构成的耦联分子对,能够进行细胞间信号转导、诱导细胞的定向迁移,参与细胞的多种生物学过程。研究证实,SDF-1/CXCR4信号轴在MSCs参与心肌缺血、肾脏病变、骨组织损伤等损伤组织修复过程中有重要的促趋化和增殖的作用。本文简要介绍了SDF-1和CXCR4的分子结构,重点阐述了SDF-1/CXCR4信号轴在MSCs参与相关损伤组织修复中的作用,归纳总结了该领域的研究进展,并展望了该领域未来的发展方向,为深入理解SDF-1/CXCR4信号轴及其在MSCs参与组织损伤修复过程中的作用提供理论基础,也为临床上更好地将MSCs应用于损伤组织修复提供参考。 Bone marrow-derived mesenchymal stem cells(MSCs)have a self-renewal capacity and multilineage differentiation potential,and play an important role in the repair of injured tissue.Stromal cell-derived factor-1(SDF-1)/CXC chemokine receptor 4(CXCR4)signal axis is a coupled molecular pair formed by the interaction between SDF-1 and CXCR4,which can carry out signal transduction,induce cell migration,and participate in a variety of biological processes of cells.Studies have confirmed that SDF-1/CXCR4 signal axis plays a pivotal role in promoting chemotaxis and proliferation in MSCs-mediated tissue repair of myocardial ischemia,kidney disease,and bone tissue injury and so on.This review paper briefly introduces the molecular structure of SDF-1 and CXCR4,then discusses the role of SDF-1/CXCR4 signal axis in MSCs-mediated repair of related injured tissue.Finally,we summarize the research progress and prospect the future development directions in this field.This review provides a theoretical basis for better understanding of SDF-1/CXCR4 axis and its role in MSCs-mediated tissue repair,and brings a reference for better application of MSCs in tissue repair in clinic.
