鸡血藤在大鼠体内的排泄动力学研究

摘要 目的:研究大鼠灌胃鸡血藤水提取物的排泄动力学,为其临床应用提供科学依据。方法:给雄性SD大鼠灌胃鸡血藤水提取物(30 g·kg^(–1),以生药量计),分别在给药后的2、4、6、8、12、24、48 h收集每组的粪便和尿液。采用超快速液相色谱-质谱法检测大鼠排泄物样品,通过与对照品比对后鉴定和定量鸡血藤水提取物中化合物。结果:在大鼠48 h排泄的尿液和粪便中可检测到20个原型化合物,即烟酸(1)、对羟基苯甲酸(2)、水杨酸(3)、6,9-二羟基大柱香波龙-4,7-二烯-3-酮(4)、大豆苷(5)、8,9-二羟基大柱香波龙-4,6-二烯-3-酮(6)、异落叶松脂素(7)、芒柄花素(8)、4′,8-二甲氧基-7-O-β-D-吡喃葡萄糖基异黄酮(9)、(+)-松脂酚(10)、(+)-表松脂酚(11)、8-甲雷杜辛(12)、芒柄花素钠(13)、3′-甲氧基大豆素(14)、鸡血藤黄醇A (15)、毛蕊异黄酮(16)、艳紫铆素A (17)、奥刀拉亭(18)、鹰嘴豆芽素A (19)和(6a R,11a R)-高丽槐素(20)。结论:鸡血藤中的一些原型化合物可通过大鼠尿液和粪便排出体外。 Objective:To investigate the excretion kinetics of the constituents in aqueous extracts of Spatholobi Caulis after oral administration to rats,and to provide a scientific basis for its clinical application.Methods:Male Sprague-Dawley(SD)strain rats were selected and intragastrically administrated with aqueous extracts of Spatholobi Caulis(equivalent to 30 g·kg^(–1) of crude Spatholobi Caulis weight).Fecal and urine samples were collected at 2,4,6,8,12,24,48 h post-dosing ultra performance liquid chromatography-tandem mass spectrometry(UFLC-MS/MS)was employed to detect compounds in rat excreta samples,and the identified and quantified compounds in Spatholobi Caulis water extract were determined through comparison with reference substances.Results:In the excreted urine and feces during 24 h post-dosing,twenty prototype compounds,namely nicotinic acid(1),p-hydroxybenzoic acid(2),salicylic acid(3),6,9-dihydroxymegastigma-4,7-diene-3-one(4),daidzin(5),8,9-dihydroxymegastigma-4,6-dien-3-one(6),isolariciresinol(7),formononetin(8),4′,8-dimethoxy-7-O-β-D-glucosylisoflavone(9),(+)-epipinoresinol(10),(+)-pinoresinol(11),8-O-methylretusin(12),formononetin sodium(13),3′-methoxydaidzein(14),spasuberol A(15),calycosin(16),butesuperin A(17),odoratin(18),biochanin A(19),and(6aR,11aR)-maackiain(20)were detected and identified.Conclusion:Some prototypical compounds in Spatholobi Caulis could be excreted by urine and feces in rats.
