脯氨酸-谷氨酸-亮氨酸富集蛋白1在甲状腺癌组织的表达及临床意义

Expression and clinical significance of proline glutamate leucine enriched protein 1 in thyroid cancer tissue

摘要 目的探讨脯氨酸-谷氨酸-亮氨酸富集蛋白1在甲状腺癌组织中的变化, 及其表达水平与肿瘤增殖和转移的关系。方法选取2018年8月至2023年8月新乡医学院第一附属医院手术治疗的44例甲状腺癌临床样本作为研究对象, 对应得癌旁组织作为对照, 采用蛋白质免疫印迹分析癌旁组织和甲状腺癌组织中脯氨酸-谷氨酸-亮氨酸富集蛋白1蛋白的表达水平。同时分析细胞增殖标志物(细胞增殖抗原)、迁移标志物(基质金属蛋白酶)和上皮-间充质转化标志物(N-钙黏蛋白和波形蛋白)表达水平。并分析脯氨酸-谷氨酸-亮氨酸富集蛋白1敲降对甲状腺癌细胞增殖、迁移和侵袭的影响。组间计量数据采用t检验。结果癌旁组织脯氨酸-谷氨酸-亮氨酸富集蛋白1表达水平(0.93±0.11)明显低于甲状腺癌组织(2.02±0.28), 差异有统计学意义(t=24.370, P<0.05)。癌旁组织细胞增殖抗原蛋白表达水平(1.01±0.29)明显低于甲状腺癌组织(2.48±0.12), 差异有统计学意义(t=19.070, P<0.05)。癌旁组织基质金属蛋白酶13表达水平(0.71±0.10)明显低于甲状腺癌组织(1.53±0.19), 差异有统计学意义(t=24.910, P<0.05)。甲状腺癌组织N-钙黏蛋白和波形蛋白表达水平(2.38±0.18、2.35±0.17)明显著高于癌旁组织(1.47±0.23、1.28±0.16), 差异有统计学意义(t=20.790、30.130, P<0.05)。脯氨酸-谷氨酸-亮氨酸富集蛋白1表达水平与细胞增殖抗原、基质金属蛋白酶13、N-钙黏蛋白和波形蛋白表达水平呈正相关(r=0.142、0.251、0.316、0.482, P<0.05)。对照组细胞吸光度值(2.04±0.14)显著高于PELP1 KD组细胞(1.51±0.10), 差异有统计学意义(t=20.790, P<0.05)。对照组迁移能力[(85.46±4.85)%]显著高于PELP1 KD组细胞[(68.84±8.69)%]差异有统计学意义(t=4.723, P<0.05)。对照组细胞侵袭数量[(142.75±14.56)个]显著高于PELP1 KD组细胞[(78.25±12.27)个], 差异有统计学意义(t=9.583, P<0.05)。结论甲状腺癌组织中脯氨酸-谷氨酸-亮氨酸富集蛋白1表达水平显著增加, 下调脯氨酸-谷氨酸-亮氨酸富集蛋白1可抑制甲状腺癌细胞的增殖、迁移和侵袭等过程。 Objective To investigate the changes of proline glutamic acid and leucine rich protein 1(PELP1)in thyroid cancer tissue and correlation between PELP1 expression level and tumor proliferation and metastasis.Methods A total of 45 clinical samples of thyroid cancer treated with surgery in the First Affiliated Hospital of Xinxiang Medical College from August 2018 to August 2023 were selected as the research subjects,and corresponding adjacent cancerous tissues were used as controls.The expression level of PELP1 protein in adjacent cancerous tissues and thyroid cancer tissues was analyzed by Western blotting.Simultaneously the expression levels of cell proliferation marker(PCNA),migration markers(MMP-1,MMP-13),and epithelial mesenchymal transition markers(N-cadherin and vimentin)were detecvted.The ability of proliferation,migration,and invasion in control cells and PELP1 KD cells was analyzed using CCK-8 assay,scratch assay,and Transwell assay.The intergroup measurement data were compared by thet-test.Results The expression level of PELP1 protein in adjacent tissues(0.93±0.11)was significantly lower than that in thyroid cancer tissues(2.02±0.28,t=24.370,P<0.05).The expression level of PCNA protein in adjacent tissues(1.01±0.29)was significantly lower than that in thyroid cancer tissues(2.48±0.12,t=19.070,P<0.05).The expression level of MMP-13 protein in adjacent tissues(0.71±0.10)was significantly lower than that in thyroid cancer tissues(1.53±0.19,t=24.910,P<0.05).The protein expression levels of N-cadherin and vimentin in adjacent tissues(1.47±0.23 and 1.28±0.16)were significantly lower than those in thyroid cancer tissues(2.38±0.18 and 2.35±0.17,t=20.790,30.130,P<0.05).The expression level of PELP1 protein was positively correlated with the expression levels of PCNA,MMP-13,N-cadherin,and vimentin proteins(r=0.142,0.251,0.316,0.482,P<0.05).The absorbance value of the control group cells(2.04±0.14)was significantly higher than that of the PELP1 KD group cells(1.51±0.10,t=20.790,P<0.05).The migration ability of the control group cells[(85.46±4.85)%]was significantly higher than that of the PELP1 KD group cells[(68.84±8.69)%,t=4.723,P<0.05].The number of invasive cells in the control group(142.75±14.56)was significantly greater than that in the PELP1 KD group(78.25±12.27,t=9.583,P<0.05).Conclusion The expression level of PELP1 protein significantly increases in thyroid cancer tissue,and downregulating PELP1 can suppress the ability of proliferation,migration,and invasion in thyroid cancer cells.
