紫苏籽蛋白提取工艺优化及抗疲劳活性研究

摘要 紫苏籽富含蛋白质和氨基酸,具有较高的开发价值,该研究利用单因素试验和正交试验对紫苏籽蛋白的提取工艺进行研究和优化,研究结果表明,紫苏籽蛋白的最佳提取工艺为料液比例1∶20、碱提pH 10、碱提时间20 min和碱提温度50℃,此时紫苏籽蛋白提取率最高,为54.25%。通过Design-Expert 12软件对正交试验数据进行多元回归拟合分析,获得紫苏籽蛋白提取工艺参数的回归模型为Y=53.12+0.45A+1.19B+1.39C-0.051AB+1.61AC-1.37BC-2.41A~2-0.81B~2-2.21C~2,此回归模型的P值<0.001,说明试验值与真实值之间的误差极小。此外,还对紫苏籽蛋白的抗疲劳生物活性进行了研究,结果表明适量的紫苏籽蛋白具有较好的抗疲劳活性。 Perilla seed is rich in protein and amino acids,and has high development value.In this study,single factor test and orthogonal test are used to study and optimize the extraction process of perilla seed protein.The research results show that the optimal extraction process of perilla seed protein is solid-liquid ratio of 1∶20,alkaline extraction pH of 10,alkaline extraction time of 20 min and alkaline extraction temperature of 50℃.At this time,the extraction rate of perilla seed protein is the highest of 54.25%.Through the multiple regression fitting analysis of orthogonal test data by Design-Expert 12 software,the regression model of perilla seed protein extraction process parameters is obtained as Y=53.12+0.45 A+1.19 B+1.39 C-0.051 AB+1.61 AC-1.37 BC-2.41 A 2-0.81 B 2-2.21 C 2,the P value of this model is<0.001,and the error between the test value and the true value is very small.In addition,the anti-fatigue biological activity of perilla seed protein is also studied,and the results show that the appropriate amount of perilla seed protein has good anti-fatigue activity.
