衰老促进主动脉瘤发生的遗传机制

摘要 主动脉瘤是以主动脉局部出现不可逆的永久扩张为特征的一类主动脉疾病。主动脉瘤发病机制复杂,与遗传和增龄密切相关。目前已发现与主动脉瘤相关的基因有超过1000个。一方面,一部分致病或者易感基因在模式生物上得到证实;另一方面,60岁以上人群中主动脉瘤的发病率急剧上升。衰老是否影响主动脉瘤相关基因,促进主动脉瘤的发生,尚不十分清楚。细胞衰老导致的线粒体功能障碍、氧化应激增加及衰老相关分泌表型增加等改变均可促进主动脉瘤的发生。该文重点综述了细胞衰老影响主动脉瘤相关基因及其机制,探讨通过延缓衰老预防主动脉瘤发生的可能性。 Aortic aneurysm is a type of aortic disease characterized by irreversible permanent dilation of the aorta.Epidemiologically genetic components and advanced ages are identified as critical risk factors for aortic aneurysm.By far,more than one thousand of genes have been associated with the disease.On the other hand,the incidence of aortic aneurysm is sharply increased with ages.However,whether aging shift the expression of these genes contributes to the development of aortic aneurysm remains largely unknown.Here we focus on searching for aortic aneurysm-associated genes that are linked to cellular senescence,discussing the possible mechanisms how aging increases the susceptibility of aortic aneurysm via changing these genes.This review provides a notion whether aortic aneurysm can be prevented by the slowdown of aging process.
