生殖支原体MG_095蛋白的生物信息学分析及抗原表位预测

摘要 目的本研究旨在应用生物信息学方法对生殖支原体MG_095蛋白的理化性质进行分析,并预测其结构和表位,以探讨其作为潜在疫苗候选抗原的可能性。方法首先,利用DEG数据库分析MG_095基因是否为生殖支原体必需基因;其次,采用Prot-Param、ProtScale、BUSCA、SOPMA、DeepTMHMM、SignalP 6.0、Motif Scan、STRING等在线软件预测生殖支原体MG_095蛋白的氨基酸组成、理化性质、亲/疏水性、亚细胞定位、二级结构、跨膜区、信号肽、修饰位点和互作蛋白;最后,使用IEDB、ABCpred、SYFPEITHI和NetMHCIIpan 4.1软件预测靶蛋白的B、T细胞抗原表位。结果靶蛋白MG_095为生殖支原体必需基因MG_095所编码,是由398个氨基酸构成的稳定亲水性蛋白,其分子式为C_(2018)H_(3127)N_(527)O_(632)S_(6),分子质量为44.93154 ku,理论等电点为8.80。该蛋白的二级结构主要由α-螺旋和无规卷曲组成,其亚细胞定位于细胞外,无跨膜区,含有信号肽,存在19个磷酸化位点、4个糖基化位点和2个N-豆蔻酰化位点,并与多个核糖体蛋白有互作关系。此外,靶蛋白MG_095含6个优势B细胞抗原表位和3个优势T细胞抗原表位。结论假定表面暴露分子MG_095具有良好的免疫原性,可作为生殖支原体潜在的疫苗候选分子。 Objective The physicochemical properties,structure and epitopes of MG_095 protein of Mycoplasma genitalium was analyzed by bioinformatics methods,which contributed to further explore its potential as a potential vaccine candidate antigen.Methods Firstly,the DEG database was used to investigate whether the MG_095 gene is an essential gene for M.genitalium;Secondly,the amino acid composition,physicochemical properties,hydrophilic/hydrophobicity,subcellular localization,secondary structure,transmembrane structural region,signal peptide,modification sites and interacting proteins of target protein MG_095 were analyzed by bioinformatics softwares,such as Prot-Param,ProtScale,DeepTMHMM,SignalP6.0,BUSCA,SOPMA,Motif Scan and STRING;Finally,B-and T-cell antigenic epitopes of target protein were predicted using software IEDB,ABCpred,SYFPEITHI and NetMHCIIpan 4.1.Results The target protein MG_095,encoded by the M.genitalium essential gene MG_095,is a stable hydrophilic protein consisting of 398amino acids,with a molecular formula of C_(2018)H_(3127)N_(527)O_(632)S_(6),a molecular mass of 44.93154ku,and a theoretical isoelectric point of 8.80.Its secondary structure consists mainly ofα-helices and random convolutions.MG_095 protein is localized to the extracellular,which contains a signal peptide??/Pro,has no transmembrane region,possesses 19 phosphorylation sites,4glycosylation sites and 2N-cardamoylation sites,and interacts with several ribosomal proteins.In addition,the target protein MG_095contains 6dominant B-cell antigenic epitopes and 3dominant T-cell antigenic epitopes.Conclusion MG_095protein,as a putative surface-exposed molecule,has excellent immunogenicity and could be a potential vaccine candidate molecule for M.genitalium.
