摘要
采用色谱柱C_(18)[Gensial C_(18),5 mm,250 mm×4.6 mm,GAt.No.GC1852546],流动相是甲醇∶水=10∶90,流速0.200 mL/min,检测波长254 nm,PDA检测器,建立一个龙眼核多酚高效液相色谱(HPLC)测定方法。样品经甲醇溶解,0.45μm有机滤膜过滤,可知多酚0.5~5μg/μL范围内线性方程为y=100738x,相关系数R=0.9998,呈良好的线性关系,平均回收率为97.8%,相对标准偏差RSD=0.00%。该方法简便快速,操作稳定,重现性好,不仅适用于龙眼核中多酚测定,而且为多酚产品中多酚含量的测定提供技术参考。
The determination was performed on C_(18)column[Gensial C_(18)5 mm 250 mm×4.6 mm GAt.No.GC1852546]with the mobile phase consisted of methanol∶water=10∶90 at the flow rate of 0.200 mL/min.The detection wavelength was 254 nm with PDA detector.A high performance liquid chromatography(HPLC)method for the determination of longan nuclear polyphenols was established.After the samples were dissolved by methanol and filtered by 0.45μm organic filter membrane,the linear equation of polyphenols in the range of 0.5~5μg/μL was y=100738x,the correlation coefficient R was 09998,showing a good linear relationship,and the average recovery rate was 97.8%.Relative standard deviation RSD was 0.00%.The method was simple,rapid,stable and reproducible.It not only was suitable for the determination of polyphenols in longan nucleus,but also provided a technical reference for the determination of polyphenols in polyphenol products.
