摘要
目的探讨LGR4基因在宫颈鳞癌增殖中发挥的作用及分子机制。方法利用小干扰siRNA(包括siLGR4和siNC)转染宫颈鳞癌Siha和Caski细胞,采用RT-qPCR技术检测其干扰效果,采用MTT检测细胞的增殖情况,采用Western blot检测宫颈鳞癌细胞Wnt/β-catenin信号通路及下游周期因子的表达情况。结果siLGR4的干扰效果较好,MTT实验提示LGR4干扰后可抑制宫颈鳞癌细胞增殖能力,Western blot实验证实LGR4干扰后可抑制Wnt/β-catenin信号通路及下游周期因子的表达。结论LGR4可通过调控Wnt/β-catenin信号通路及下游周期因子促进宫颈鳞癌细胞增殖,有望成为宫颈鳞癌的潜在分子标志物及治疗靶点。
Objective To explore the role and molecular mechanism of LGR4 gene in the proliferation of squamous cervical cancer(SCC).Methods SCC Siha and Caski cells were transfected with small interfering siRNAs(including siLGR4 and siNC),and the interfering effects were detected by RT-qPCR,the proliferation of the cells was detected by MTT,and the expression of Wnt/β-catenin signaling pathway and downstream cyclic factors of the cervical squamous carcinoma cells was detected by Western blot.Results The interference effect of siLGR4 was better.And MTT assay suggested that the proliferation ability of cervical squamous carcinoma cells in siLGR4 group could be inhibited after LGR4 interference,and Western blot assay confirmed that the expression of Wnt/β-catenin signaling pathway and downstream cyclic factors could be inhibited after LGR4 interference.Conclusions LGR4 can promote the proliferation of cervical squamous carcinoma cells by regulating the Wnt/β-catenin signaling pathway and downstream cyclic factors,and is expected to be a potential molecular marker and therapeutic target for cervical squamous carcinoma.
