摘要
以花生网斑病的病原菌花生派伦霉为材料,选择对戊唑醇的抗性菌株与敏感菌株,比较两者对渗透压的敏感性,以及在戊唑醇胁迫下膜透性的差异,重点探讨抗戊唑醇菌株的生理生化特点。结果表明,随着试验设置的NaCl浓度的增加,培养环境渗透压的升高,敏感菌株和抗性菌株的菌落直径均表现先升后降的趋势。敏感菌株对低渗透压环境不敏感,对中、高渗透压环境较为敏感;抗性菌株对NaCl低、中渗透压环境均不敏感,只对高渗透压环境敏感。经不同浓度戊唑醇处理后,供试菌株的相对渗透率均随时间增加呈上升趋势,敏感菌株的相对渗透率低于同期的抗性菌株,说明后者的细胞膜通透能力较强,对戊唑醇的排泄能力也较强,而敏感菌株的细胞膜通透能力较差,对戊唑醇反应敏感。试验结果可为花生网斑病菌对戊唑醇抗性机制的研究提供相关依据。
In this paper,the osmotic pressure sensitivity and membrane permeability of tebuconazole-resistant and sensitive strains of Peyronellaea arachidicola,pathogen of peanut web blotch disease,were compared,focusing on the physiological and biochemical characteristics of tebuconazole-resistant strains.The results indicated that under different concentrations of NaCl,both sensitive and resistant strains showed an initial increase in colony diameter followed by a decrease.Sensitive strains were less affected by low osmotic pressure but were more sensitive to medium and high osmotic pressure environments.In contrast,resistant strains were not sensitive to low and medium osmotic pressure environments,but were sensitive to high osmotic pressure environments.Following treatment with varying concentrations of tebuconazole,the relative permeability of the test strains increased over time to a certain extent.The relative permeability of S-TL-11 and S-JL-20 were lower than those of R-AS-02 and R-LY-72 under each concentration of chemical treatment.It showed that the resistant strains had stronger cell membrane permeability and can excrete more tebuconazole,while the sensitive strains had poorer cell membrane permeability and were more sensitive to tebuconazole.The results will provide a theoretical basis for further exploration of the resistance mechanism of P.arachidicola to tebuconazole and guide the scientific use of fungicides in the field.
